Multifunctional PLGA particles containing poly(l-glutamic acid)-capped silver nanoparticles and ascorbic acid with simultaneous antioxidative and prolonged antimicrobial activity

AB  - A water-soluble antioxidant (ascorbic acid, vitamin C) was encapsulated together with poly(l-glutamic acid)-capped silver nanoparticles (AgNpPGA) within a poly(lactide-co-glycolide) (PLGA) polymeric matrix and their synergistic effects were studied. The PLGA/AgNpPGA/ascorbic acid particles synthesized by a physicochemical method with solvent/non-solvent systems are spherical, have a mean diameter of 775 nm and a narrow size distribution with a polydispersity index of 0.158. The encapsulation efficiency of AgNpPGA/ascorbic acid within PLGA was determined to be >90%. The entire amount of encapsulated ascorbic acid was released in 68 days, and the entire amount of AgNpPGAs was released in 87 days of degradation. The influence of PLGA/AgNpPGA/ascorbic acid on cell viability, generation of reactive oxygen species (ROS) in HepG2 cells, as well as antimicrobial activity against seven different pathogens was investigated. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay indicated good biocompatibility of these PLGA/AgNpPGA/ascorbic acid particles. We measured the kinetics of ROS formation in HepG2 cells by a DCFH-DA assay, and found that PLGA/AgNpPGA/ascorbic acid caused a significant decrease in DCF fluorescence intensity, which was 2-fold lower than that in control cells after a 5 h exposure. This indicates that the PLGA/AgNpPGA/ascorbic acid microspheres either act as scavengers of intracellular ROS and/or reduce their formation. Also, the results of antimicrobial activity of PLGA/AgNpPGA/ascorbic acid obtained by the broth microdilution method showed superior and extended activity of these particles. The samples were characterized using Fourier transform infrared spectroscopy, field-emission scanning electron microscopy, transmission electron microscopy, zeta potential and particle size analysis. This paper presents a new approach to the treatment of infection that at the same time offers a very pronounced antioxidant effect.

AB  - Silver nanoparticles (AgNps) were prepared by modified chemical reduction with poly (α, γ, l-glutamic acid) (PGA) as capping agent. These Ag/PGA nanoparticles (AgNpPGAs) were highly stable over long periods of time without signs of precipitation. In addition to obtaining stable AgNpPGAs, a further aim was to examine their encapsulation in the poly(L-lactide-co-glycolide) (PLGA) polymer matrix. The current interest of polymer-AgNps in biomedical applications is because a versatile system must have antimicrobial activity upon target contact, without the release of toxic biocides. The synthesis of these PLGA/AgNpPGAs used physicochemical methods with solvent/non-solvent systems. Degradation of these PLGA/AgNpPGAs and the release rate of their AgNPs were studied in physiological solution over three months. The antimicrobial activity of the samples was investigated towards six laboratory control strains from the American Type Culture Collection (ATCC) and one clinical isolate methicillin-resistant Staphylococcus aureus strain by the broth microdilution method and the results showed superior and extended activity of PLGA/AgNpPGAs. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay indicated good biocompatibility of these PLGA/AgNpPGAs. The formation of intracellular reactive oxygen species was measured spectrophotometrically using a fluorescent probe, which showed that these PLGA/AgNpPGAs are not inducers of such species. The samples were characterized by UV–VIS spectrometry, X-ray diffraction, zeta potential measurements, field-emission scanning electron microscopy, and transmission electron microscopy.

AB  - A simple and green chemical method has been developed to synthesize stable bare and capped silver nanoparticles based on the reduction of silver ions by glucose and capping by poly(α,γ,L-glutamic acid) (PGA). The use of ammonia during synthesis was avoided. PGA has had a dual role in the synthesis and was used as a capping agent to make the silver nanoparticle more biocompatible and to protect the nanoparticles from agglomerating in the liquid medium. The synthesized PGA-capped silver nanoparticles in the size range 5–45 nm were stable over long periods of time, without signs of precipitation. Morphological examination has shown that the silver nanoparticles had a nearly spherical, multiply twinned structure. The effects of the reaction temperature and the reaction time during the synthesis were investigated too. The biocompatibility of the PGA-capped silver nano-particles is discussed in terms of in vitro toxicity with human intestinal Caco-2 cells. The samples were characterized by UV–Visible spectroscopy, field emission scanning electron microscopy, transmission electron microscopy, and zeta potential measurements.

AB  - It is well known that selenium is an essential trace element playing the important roles in mammalian biology. For years, its organic forms and some salts have been used in studying its biological effects, but recently, selenium nanoparticles have gained great attention as a possible material for cancer therapy and prevention. In this study we report the facile and reproducible method for synthesis the nanoselenium by reduction of sodium selenite with an ascorbic acid, in the presence of a two stabilizing agents, bovine serum albumin (BSA) or poly (L-glutamic acid) (PGA). The influence of the sodium selenite/ascorbic acid ratio as well as the types and concentration of two different stabilizers on the wide variety physicochemical characteristics of obtained particles were examined.

AB  - ZnO represent one of the most important multifunctional materials. Its properties are well known and confirmed in different application areas such as: gas sensors, UV lasers, solar cells, electroluminescent and optoelectronic devices, piezoelectric transducers, hydrogen-storages, rubber industry and plastic processing, cosmetics and pharmacy, and it is also used as an antibacterial agent. In this work, we propose a low-temperature hydrothermal method for the synthesis of phase-pure ZnO powders with a controlled morphology and narrow particle size distribution. This simple and low-cost method allows tailoring of the shape and size of ZnO particles, from micro-rods via hexagonal prism-like to nano-spheres, by choosing the appropriate pH of the reaction solution, tuned by the varying of [Na+]:[Zn2+] molar ratio. The agglomeration of the powders was prevented with the assistance of PVP as a capping agent. The synthesized powders were characterized by XRD, TEM, SEAD and HRTEM methods to examine the phase purity and crystallinity. FE-SEM measurements were used for the morphology characterization, while the results of Raman and UV-Vis DRS measurements were used for the study of optical properties. The relationship between the particle size and morphology and the optical properties is discussed. Diffuse reflectance spectra of ZnO powders with different size and morphology revealed characteristic R curves with the absorption edge near 380 nm, but with obvious difference in the absorption in the visible region. The micro-sized powder ZnO8 revealed the lowest reflectance (∼70%), compared to submicro- (∼80%) and nano-sized (ZnO12 and ZnO13, ∼90%) powders. Thus, the reflectance of the ZnO powders decreases with the increase in the average particle size.

AB  - In this paper, we investigate the effect of the particle size and morphology on the optical properties of ZnO. A series of ZnO micro and nanocrystals were synthesized by the hydrothermal processing of zinc acetate dihydrate and sodium hydroxide as the starting materials, and polyvinylpyrrolidone (PVP) as the polymer surfactant. The particle size and morphology were tailored by adjusting the reactant molar ratios [Zn2+]/[OH−], while the reaction temperature and the time remained unchanged. X-ray diffraction (XRD), transmission electron microscopy (TEM), selected area electron diffraction (SAED) and high-resolution TEM (HRTEM) have shown that the micro and nanocrystals have a high crystalline pure wurtzite-type hexagonal structure with nanosized crystallites. The size and morphology of the ZnO micro and nanocrystals were investigated by field emission scanning electron microscopy (FE-SEM), which showed a modification from micro-rods via hexagonal-faceted prismatic morphology to nanospheres, caused by simple adjustment of the reactant molar ratio [Zn2+]/[OH−] from 1:1 to 1:5. The optical properties of the ZnO micro and nanocrystals, as well as their dependence on the particle size and morphology were investigated by Raman and ultraviolet–visible (UV–vis) diffuse reflectance spectroscopy (DRS). The UV–vis spectra showed that the modification of the particle size and morphology from nanospheres to micro-rods resulted in increased absorption, and a slight red-shift of the absorption edge (0.06 eV). Besides, the band gap energy of the synthesized ZnO micro and nanocrystals showed the red shift (∼0.20 eV) compared to bulk ZnO. According to the results of a Raman spectroscopy, the enhanced visible light absorption of the ZnO micro and nanocrystals is related to two phenomena: (1) the existence of lattice defects (oxygen vacancies and zinc interstitials), and (2) the particle surface sensitization by PVP.

AB  - The great importance of hydroxyapatite in the field of biomaterial science inspires researchers to investigate various approaches to adjust and improve existing and to find out new useful properties of this class of materials. Doping of original hexagonal apatite crystal structure with a number of ions has been shown to improve phase stability, mechanical and electrical properties, as well as its biological applicability. Fabrication of Zr-HAp materials could be significant for mechanical properties improvement, teeth implant color adjustment, altogether with conserved bioactivity and without cell toxicity. In this study, simple chemical precipitation is used to synthesize zirconium-doped hydroxyapatite, Zr-HAp, with 0, 1.0, 5.0 and 10.0 at.% of Zr. Phase purity was investigated by XRD, particles morphology by electron microscopy, while middle range arrangement and presence of different functional groups through IR and Raman spectroscopy studies. Efficiency of Zr-ions incorporation is checked by EDX chemical analysis.

AB  - Phase composition, crystal structure and morphology of biological hydroxyapatite (BHAp) extracted from human mandible bone, and carbonated hydroxyapatite (CHAp), synthesized by the chemical precipitation method, were studied by x-ray powder diffraction (XRD), Fourier transform infrared (FTIR) and Raman (R) spectroscopy techniques, combined with transmission electron microscopy (TEM). Structural and microstructural parameters were determined through Rietveld refinement of recorded XRD data, performed using the FullProf computing program, and TEM. Microstructural analysis shows anisotropic extension along the [0 0 l] crystallographic direction (i.e. elongated crystallites shape) of both investigated samples. The average crystallite sizes of 10 and 8 nm were estimated for BHAp and CHAp, respectively. The FTIR and R spectroscopy studies show that carbonate ions substitute both phosphate and hydroxyl ions in the crystal structure of BHAp as well as in CHAp, indicating that both of them are mixed AB-type of CHAp. The thermal behaviour and carbonate content were analysed using thermogravimetric and differential thermal analysis. The carbonate content of about 1 wt.% and phase transition, at near 790 °C, from HAp to β-tricalcium phosphate were determined in both samples. The quality of synthesized CHAp powder, particularly, the particle size distribution and uniformity of morphology, was analysed by a particle size analyser based on laser diffraction and field emission scanning electron microscopy, respectively. These data were used to discuss similarity between natural and synthetic CHAp. Good correlation between the unit cell parameters, average crystallite size, morphology, carbonate content and crystallographic positions of carbonate ions in natural and synthetic HAp samples was found.

AB  - A series of cobalt-exchanged hydroxyapatite (CoHAp) powders with different Ca/Co ratios and nominal unit-cell contents Ca10-x Cox (PO4)6(OH)2, x = 0, 0.5, 1.0, 1.5 and 2.0, were synthesized by hydrothermal treatment of a precipitate at 473 K for 8 h. Based on ICP (inductively coupled plasma) emission spectroscopy analysis, it was established that the maximum amount of cobalt incorporation saturated at 7sim;12 at.% under these conditions. The effects of cobalt content on the CoHAp powders were investigated using ICP emission spectroscopy, particle size analysis, transmission electron microscopy (TEM) and high-resolution transmission electron microscopy (HRTEM) analyses as well as X-ray powder diffraction (XRPD) including Rietveld analysis. According to XRPD, all the materials are single-phase HAp and CoHAp of low crystallinity. Rietveld analysis shows that Co enrichment causes the c cell parameter to decrease at a faster rate than the a cell parameter. A microstructural analysis showed anisotropic X-ray line broadening due to crystallite size reduction. In CoHAp there is significant crystal elongation in [001], and the average size decreases with increasing cobalt content. The crystallite morphology transforms from rod-like for the pure HAp to lamellae at the highest degree of Co substitution. The results of Rietveld refinement (symmetry, size and morphology of the crystallites) were confirmed by TEM and HRTEM analysis. © 2010 International Union of Crystallography. AB  - Fabrication of full dense ceramic materials on the basis of hydroxyapatite (HAp) and biphasic calcium phosphate (BCP) with controllable microstructural and compositional characteristics attracts considerable efforts. Various synthesis and sintering methods were applied in order to achieve desirable material properties. In this study different nanopowders were produced and processed via two-step sintering (TSS) approach. Characterization of synthesized nanopowders were done by XRD, BET, FE-SEM, TEM and thermal analysis methods, while microstructural and chemical characterizations of sintered samples were performed through FE-SEM and XRD analysis. A possibility for obtaining full dense ceramics with suppressed grain growth is discussed on the basis of inherent nanopowders characteristics. Certain attention would be paid on thermal behavior of Ca-deficient HAp systems.
