On the Role of Poly-Glutamic Acid in the Early Stages of Iron(III) (Oxy)(hydr)oxide Formation

AB  - Nucleation of minerals in the presence of additives is critical for achieving control over the formation of solids in biomineralization processes or during syntheses of advanced hybrid materials. Herein, we investigated the early stages of Fe(III) (oxy)(hydr)oxide formation with/without polyglutamic acid (pGlu) at low driving force for phase separation (pH 2.0 to 3.0). We employed an advanced pH-constant titration assay, X-ray diffraction, thermal analysis with mass spectrometry, Fourier Transform infrared spectroscopy, and scanning electron microscopy. Three stages were observed: initial binding, stabilization of Fe(III) pre-nucleation clusters (PNCs), and phase separation, yielding Fe(III) (oxy)(hydr)oxide. The data suggest that organic–inorganic interactions occurred via binding of olation Fe(III) PNC species. Fourier Transform Infrared Spectroscopy (FTIR) analyses revealed a plausible interaction motif and a conformational adaptation of the polypeptide. The stabilization of the aqueous Fe(III) system against nucleation by pGlu contrasts with the previously reported influence of poly-aspartic acid (pAsp). While this is difficult to explain based on classical nucleation theory, alternative notions such as the so-called PNC pathway provide a possible rationale. Developing a nucleation theory that successfully explains and predicts distinct influences for chemically similar additives like pAsp and pGlu is the Holy Grail toward advancing the knowledge of nucleation, early growth, and structure formation.

Permanent disposal of Cs ions in the form of dense pollucite ceramics having low thermal expansion coefficient

AB  - A promising method for removal of Cs ions from water and their incorporation into stable crystal structure ready for safe and permanent disposal was described. Cs-exchanged X zeolite was hot-pressed at temperature ranging from 800 to 950 °C to fabricate dense pollucite ceramics. It was found that the application of external pressure reduced the pollucite formation temperature. The effect of sintering temperature on density, phase composition and mechanical properties was investigated. The highest density of 92.5 %TD and the highest compressive strength of 79 MPa were measured in pollucite hot-pressed at 950 °C for 3 h. Heterogeneity of samples obtained at 950 °C was determined using scanning electron microscopy. The pollucite hot-pressed at 950 °C had low linear thermal expansion coefficient of ∼4.67 × 10−6 K−1 in the temperature range from 100 to 1000 °C. © 2019

AB  - The influence of different milling conditions obtained using two high-energy mills on hydrogen desorption from MgH2-WO3 composites was investigated. The morphology, particle and crystallite size were studied as a function of milling speed, vial's volume, and ball-to-powder ratio. The vial's fill level, the number, and type of milling balls and additive's content kept constant. Changes in morphology and microstructure were correlated to desorption properties of materials. Higher milling speed reduced particle size but, there is no significant crystallite size reduction. On the other hand, additive distribution is similar regardless of the energy input. It has been noticed that different energy input on milling blend, which is the result of combined effects of above-mentioned factors, reflects on desorption temperature but not on the kinetics of desorption. In fact, desorption mechanism changes from 2D to 3D growth with constant nucleation rate, despite obtained changes in microstructure or chemical composition of the material.

AB  - Nanotechnology has great potential in the biomedical field. Among other nanomaterials, inorganic nanoparticles have become extremely important since they possess unique physicochemical properties influenced by their specific surface structure. Consequently, inorganic nanoparticles exhibit enhanced functionalities such as biological response, antibacterial and antiviral properties, as well as optical, magnetic, and electrical responses. They have found applications in medicine, pharmacy, controlled drug delivery, optics, electronics, etc. In this chapter, reports on obtaining different metallic and ceramic inorganic nanoparticles such as gold, silver, selenium, copper, iron, zinc oxide, and hydroxyapatite for biomedical applications will be addressed. For each of these nanosystems, the main challenges regarding the currently achieved functional properties and further perspectives will also be presented.

AB  - The aim of this study was to improve the mechanical properties and to optimize antimicrobial activity of hydroxyapatite (HAP) by simultaneous doping with Mg and Cu ions in order to obtain material that would be able to assist in the bone/tooth healing process, prevent post-implementation infections and provide satisfying values of hardness and fracture toughness for biomedical application. Ion doping was done during the hydrothermal synthesis of HAP powders, whereby the content of Mg ions in the starting solution was varied between 1-20 mol. % with regard to Ca ions, while the amount of Cu ions was kept constant at 0.4 mol. %. The green compacts were sintered for 2 h at temperatures ranging 750–1200 °C depending on the Mg content, chosen in agreement with dilatometry results. Presence of Mg ions was found to favour transition from HAP to β−tricalcium phosphate phase (β−TCP), which enabled formation of biphasic HAP/β−TCP and pure β−TCP phase at 160 °C during hydrothermal synthesis. In vitro investigation of antimicrobial activity against Escherichia coli, Staphylococcus aureus and Enterococcus faecalis showed satisfactory antimicrobial activity. MTT assay performed on MRC-5 and L929 cell lines showed excellent cytocompatibility and cell proliferation. Maximum hardness by Vickers and fracture toughness values, 4.96 GPa and 1.75 MPa m1/2 respectively, were obtained upon addition of 5 mol. % Mg, as a consequence of the lowest grain size and porosity, as well as the highest densification rate. This is, to the best of our knowledge, the highest fracture toughness for HAP or β-TCP ceramics reported thus far.

The reaction mechanism that assumes the initial interaction between phosphates from LFP and calcium from HAp was proposed, generating calcium vacancies and contributing to HAp densification. Cross-sections of sintered samples showed changes in microstructural properties, with uniform atomic distribution and presence of Li2Fe3O4 spherical inclusions (200 nm) located at grain boundaries of calcium phosphate matrix. The Rietveld refinement analysis indicated changes in structural and microstructural parameters like crystallite size, anisotropy and microstructural strain of HAp upon LFP addition. Mechanical characterisation indicated improvements in fracture behaviour upon LFP addition.

AB  - Magnesium hydride, as hydrogen storage material, meets the requirements such as high gravimetric hydrogen capacity (7,6 wt%), low cost and weight, abundance and H2 absorption/desorption cycling possibility. Given that the oxide additives show the good impact on desorption properties, mechanical milling of MgH2 with addition of 5, 10 and 15% wt. WO3 was performed. The microstructure and morphology of composites were analysed by XRD, PSD and SEM and correlated to hydrogen desorption properties which have been investigated by DSC. The results have shown that WO3 has a positive effect on the desorption kinetics as well as on the change of the desorption mechanism.

AB  - The densification behavior of hydroxyapatite nanorods prepared by chemical precipitation method in open reactor conditions was investigated by application of different heating rates. The non-isothermal processing was performed with 2, 10, and 50 °C/min up to 1200 °C and yielded fully dense ceramics. The implementation of the higher heating rate provided grain size refinement from micrometer level for the slowest ramp, down to 250 nm in the case of processing with 50 °C/min, without any drawbacks regarding final density. The relative amount of retained structural hydroxyl groups in sintered ceramics was gradually increased with the heating rate. Furthermore, the qualitative level of optical translucency was increased with a higher heating rate which can be explained by the beneficial alignment of HAp nanorods during the fast heating rate processing, achieved microstructural refinement, and higher amount of structural hydroxyl groups.

AB  - Sintering of hydroxyapatite ceramics has been a challenging topic for decades since the material has good bioactivity, proven biocompatibility, low cost, and high availability. Besides its biomaterial applications, hydroxyapatite has been used so far as a catalyst support, sensor material, etc. For improvements in its functional properties, new synthesis and processing routes are certainly required. This work will present current achievements in new processing routes of hydroxyapatite ceramics. In the first part, sintering of hydroxyapatite in the presence of lithium iron phosphate will be presented. Such composition induces formation of liquid phase during sintering and interaction between materials that provides decreasing of the processing temperature and formation of reinforcing Fe-rich phase located along the grain boundaries of the matrix material. Furthermore, an influence of heating rate on pure hydroxyapatite sintering will be presented showing that conventional processing with high heating rates can be beneficial for microstructural refinement without any drawbacks regarding the final density of sintered ceramics. This will be discussed regarding the chemical changes induced due to release of hydroxyapatite structural ionic species.

AB  - In this paper, bismuth ferrite (BFO) particles synthesized by controlled hydrothermal process, where the particles of small sizes and with high purity were obtained. Structural analysis showed that non-annealed powder can be perfectly fitted to rhombohedral space group R3c and contains a very small amount of secondary phase, whereas the final product (annealed at 800 °C) represents single-phase perovskite powder with high crystallinity. HRTEM analysis confirmed existence of twin stacking faults, which are responsible for enhanced magnetic properties. EPR measurements suggested existence of electrons trapped by vacancies or defects. It has been proposed that existence of Fe3+ −OV defect complex could be generated at elevated temperatures followed by formation of trivalent Fe ions, which intensely provide local 3d moments.

Alternating current electric field modified synthesis of hydroxyapatite bioceramics

AB  - This study presents an innovative approach towards wet chemical synthesis of hydroxyapatite bioceramics by application of alternating current (AC) electric field during the synthesis at low and intermediate temperatures. Specially designed low-cost electrical setup provided stable, reliable and modular system which supplied electric energy within the reaction volume as characterized by computer simulation of reaction conditions. Energy introduced through external excitation (alternating voltage amplitude of 10 V and frequency of 1 kHz) influenced crystallization, ionic composition, sintering behavior of hydroxyapatite powders, microstructure and final phase composition of sintered ceramics. Crystallite size in [002] crystallographic direction increased with electric field assistance regardless of the synthesis temperature. Non-isothermal sintering studies showed significantly improved densification and implied better thermal stability of powders synthesized in the presence of AC field, shifting the Ca/P ratio towards stoichiometric one. Vibrational spectroscopy analysis indicated the role of charge, mobility and effective ionic radius of present ions in transferring the energy supplied with external field influencing further thermal stability of the crystal lattice. Microstructural investigation and phase composition analysis suggested that application of AC electric field during the synthesis of nanocrystalline hydroxyapatite improved material properties and offered potential for tailoring macro/micro-porosity with precise modulation of electrical parameters.

AB  - Cesium aluminosilicate phases are of the great interest as possible host for Cs immobilization in radioactive waste management. The possibility to use zeolite as a host material for radioactive Cs immobilization was investigated. Cs-exchanged forms of clinoptilolite and 13X which were prepared by ion-exchange treatment were compacted. The powders compacts of exchanged zeolites were thermally treated at 1200 °C. The XRD analysis showed that Cs was successfully immobilized after heat treatment by formation of stable cesium-aluminosilicate ceramic forms. Thermal and mechanical properties of the sintered samples were investigated. From the perspective of these characteristics, Cs-exchanged zeolite (clinoptilolite and 13 X) can be considered as a potential material for safe waste disposal.

AB  - Sintered hydroxyapatite bioceramics have been widely studied as a potential material for bone tissue reparation, however, concerning its microstructural and mechanical properties some limits were achieved at the moment. Addition of other materials that could improve functionalities, while preserving inherent advantages of this bioactive ceramics is desirable strategy. In this work, a new idea of addition of lithium iron phosphate as hydroxyapatite sintering aid, provoking liquid phase sintering in the intermediate sintering phase, has been evaluated from the point of view of thermal and dilatometric studies in inert atmosphere, with coupled mass spectroscopy monitoring. Detailed characterization of prepared materials and sintered products is given, confirming the proof of concept. Sintering ability was significantly enhanced and important microstructural features were obtained.

AB  - Copolymer poly (DL-lactide-co-glycolide) (PLGA), due of its biodegradable and biocompatible nature, is widely used in various medical applications; controlled release of delivering drugs, carriers in the tissue engineering, etc. On the other hand, zinc oxide (ZnO) is extensively used in medicine and pharmacy for personal care products, as well as in biomedical materials like dental composites, as a material for treatment of a variety of skin irritations, to enhance the antibacterial activity of different medicaments, etc. In this research we have dealt with a procedure to prepare particles of poly (lactide-co-glycolide) and nano zinc oxide (PLGA/nano-ZnO). Nano-ZnO has been synthesized using a microwave synthesis method and additionally immobilized within PLGA by physicochemical solvent/non-solvent method. Firstly, ZnO has been dispersed in acetone and then additionally added dropwise in the PLGA/ethyl acetate (PLGA/nano-ZnO(EtAc) or PLGA/acetone (PLGA/nano-ZnO(Ac)) solutions, respectively. The as-prepared dispersions were dried in air atmosphere for 24 h. 
The characterization of the prepared samples was performed using X-ray powder diffraction (XRPD) method for the structure properties, field emission scanning electron microscopy (FE SEM) for the investigation of particles morphology, as well as Malvern’s Mastersizer instrument for particle size distribution. DTA-TG measurements were performed in order to investigate the samples thermal stability and mass loss percentage. The antimicrobial behavior of the synthesized PLGA/nano-ZnO particles was tested against gram-negative and gram-positive bacteria cultures and also against Candida Albicans, diploid fungus.

During sintering procedure SHAp composition remained stable (HAp) while CDHAp was phase transformed in biphasic calcium phosphate (BCP, i.e. HAp+β-TCP) yielding HAp/BCP FGMs. To reduce mismatch stress between adjacent layers, generated during sintering, gradient of phase composition was optimized by introducing intermediate layer(s) made from a mixture of SHAp and CDHAp in an appropriate mass ratio. Optimal processing conditions for preparation of nanostructured ceramics were provided via two-step sintering. High-quality FGMs structure, without delamination, cracks and micro-structural damages was observed by an SEM technique; moreover, a detailed examination by FE-SEM established the unidirectional (perpendicular with respect to the layers) gradual change of microstructure, from fully dense to ~40% of porosity. Detailed micro-Raman and FTIR spectroscopy studies showed an increase of β-TCP amount along the FGMs height, confirming gradient of phase composition. Mechanical properties measured by microindentation revealed gradual change of microhardness from 650 to 115 HV, with simultaneous change of Young's modulus from 92 to 24 GPa. © 2014 Elsevier Ltd and Techna Group S.r.l. AB  - n the bone tissue engineering field, there is a growing interest in the application of bioactive glass scaffolds (45S5Bioglass®) due to their bone bonding ability, osteoconductivity and osteoinductivity. However, such scaffolds still lack some of the required functionalities to enable the successful formation of new bone, e.g. effective antibacterial properties. A large number of studies suggest that selenium (Se) has significant role in antioxidant protection, enhanced immune surveillance and modulation of cell proliferation. Selenium nanoparticles (SeNp) have also been reported to possess antibacterial as well as antiviral activities. In this investigation, uniform, stable, amorphous SeNp have been synthesized and additionally immobilized within spherical PLGA particles (PLGA/SeNp). These particles were used to coat bioactive glass-based scaffolds synthesized by the foam replica method. Samples were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and transmission electron microscopy (TEM). SeNp, 45S5Bioglass®/SeNp and 45S5Bioglass®/PLGA/SeNp showed a considerable antibacterial activity against Gram positive bacteria, Staphylococcus aureus and Staphylococcus epidermidis, one of the main causative agents of orthopedic infections. The functionalized Se-coated bioactive glass scaffolds represent a new family of bioactive, antibacterial scaffolds for bone tissue engineering applications.

However such scaffolds still lack some of the required functionalities to enable the successful formation of new bone, e.g. enhanced bioactivity by incorporation of bioactive molecules or growth factors and effective antibacterial properties. A large number of epidemiological, preclinical, and clinical trials suggest that selenium (Se) has significant role in antioxidant protection, enhanced immune surveillance and modulation of cell proliferation. Selenium nanoparticles (SeNp) have also been reported to possess antibacterial as well as antiviral activities. Studies also provide evidence that Se intake may be necessary for bone health. Poly(lactide-co-glycolide) (PLGA) micro and nanoparticles are used for the controlled delivery of several classes of medicaments such as growth factors, antibiotics, antimicrobial agents etc. Uniform, stable, amorphous SeNps have been synthesized and additionally encapsulated within spherical PLGA particles (PLGA/SeNps). Bioglass scaffolds have been synthesized by foam replica method and additionally coated by SeNp or by PLGA with encapsulated SeNp. Samples were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and transmission electron microscopy (TEM). SeNp, Bioglass®/SeNp and Bioglass®/PLGA/SeNp showed a considerable antibacterial activity against Gram positive bacteria Staphylococcus aureus and Staphylococcus epidermidis, one of the main causative agents of orthopedic infections.

AB  - Tissue engineering (TE) is a growing field which provides helpful alternative strategies for conventional treatments in medicine. TE involves the smart combination of cells, biomolecules and engineered porous biomaterials in the form of 3D scaffolds. When it comes to bone regeneration the use of 3D scaffolds made of calcium phosphate is a well-known concept with a great potential. Here we present the foam replica method as a procedure suitable for producing highly porous scaffolds with the pore size in the range of 100-500 μm and the mean porosity of >90%. The obtained scaffolds were further coated with selenium nanoparticles (SeNp) and SeNp immobilized within poly(epsilon caprolactone) microspheres (PCL/Se). The purpose of such coating is based on the potential anticancer activity of SeNp as well as on their prolonged release from a biodegradable polymeric carrier. Scaffolds were characterized by X-ray diffraction, scanning electron microscopy, optical microscopy, thermogravimetric/differential thermal analysis (TGA-DTA) as well as Fourier transform infrared spectroscopy (FTIR). The cytotoxicity was determined employing 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and all the samples have shown good biocompatibility. Based on these preliminary results the obtained system can be considered as a candidate for the repair of bone lesions and damages.
