AB  - Synthesis of monosized and spherical Up-Converting NanoParticles (UCNPs) with biocompatible surface are of a great interest because of their potential application in biomedicine as biomarkers or drug delivery systems. Among different synthesis routes reported in the literature, a hydro/solvo thermal method is consider to be most potential one for scaling-up due fact that is simple and economically cost-effective. In this work the synthesis of NaY0.65Gd0.15F4:Yb,Er UCNPs were performed through chitosan assistant solvothermal synthesis at 200oC. Variation of precursors concentration, type of solvent and synthesis time were performed in order to explore their influence on the structural, morphological and optical properties of the UCNPs. The XRD analysis showed that with a smaller surplus of fluoride ions the formation of Y0.65Gd0.15F4:Yb,Er orthorhombic phase occurs, while the increase of fluoride content or reaction time leads to NaY0.65Gd0.15F4:Yb,Er cubic phase formation. Along with it, the changes of UCNPs morphology from spindle to spherical shape is detected. All samples emitintense green emission due to the (2H11/2, 4S3/2) → 4I15/2electronic transitions, after been excited with infrared light (λ=978 nm).

AB  - In this work we present new up-conversion materials, consisted of SrGd2O4 matrix co-doped with different Yb3+ (1, 2.5 and 5 at%) and constant Er3+ (0.5 at%) concentrations, prepared by gycine-assisted combustion method. X-ray powder diffraction (XRPD) showed that assynthesized nanoparticles have orthorhombic structure (Pnma), assigned to the JCPDS Card No:01-072-6387. Rietveld refinement indicated a decrease in the size of the unit cell, lattice parameters, and cell volume, due to successful doping of Yb3+ and Er3+ ions into the matrix. Transmission electron microscopy (TEM) revealed that obtained nanostructure is composed of agglomerated nanoparticles, while energy dispersive spectroscopy (EDS) confirmed uniform distribution of all constituting elements in them. Up-conversion (UC) luminescence spectra measured in function of laser pumping power indicated that two-photon UC process is established in nanoparticles as a result of the trivalent erbium f-f electronic transitions: there are two green emission bands at 523 and 551 nm (2H11/2, 4S3/2 → 4I15/2) and a red emission band at 661 nm (4F9/2 → 4I15/2). The rise of Yb3+ concentration from 1 to 5 at% provokes a considerable change of the green to red ratio which indicates the possibility to optimize the color output.

Sonochemical synthesis of up-converting β-NaYF4: Yb, Er nanoparticles

AB  - Up-converting nanoparticles (UCNPs) with unique ability to convert NIR to VIS light (anti-Stokes process) have a wide application in optoelectronics, forensic, security labeling and biomedicine. Over the past few years different methods (like co-precipitation, thermal decomposition, hydro/solvothermal synthesis, etc.) are used for synthesis of β-NaYF4: Yb/Er phase, mainly from toxic organic precursors. In this work we explore conditions for stabilization of β-NaYF4: Yb/Er phase in nanoparticles applying sonochemistry synthesis of common inorganic precursor. The XRPD analysis showed that pure β phase is possible to obtained after 120 min of ultrasonification, while for shorter processing time (30-90min), cubic α-NaYF4: Yb/Er or orthorhombic YF3 : Yb,Er phase were founded too. The SEM and TEM analysis reveal presence of elongated nanoparticles self-assembled in spindles long up to 500 nm. Evolution of particle morphological and particle composition are analysed and correlated further with intensity of green emission (2H11/2,4S3/2→4I15/2) intensity measured under excitation of 978 nm.

AB  - Up-conversion is the optical property of materials which have ability to convert low energy photons (usually from infrared spectrum) into higher energy photons (from visible spectrum). Such compounds, usually lanthanide doped oxides or fluorides, have many applications in photoluminescence science and technology today. Currently, (β) NaYF4:Yb,Er polymorph is considered to be the most efficient up-converting material. Its synthesis usually includes the usage of toxic solvents, long-term heating, high pressures, controlled gas atmosphere, etc. The aim of this work was to utilize rarely applied sonochemical synthesis for the stabilization of (β) NaYF4:Yb,Er phase under mild and environmental friendly conditions. For this purpose, we performed ultrasonic treatment of common nitrate precursors and sodium fluorine for a different time. The obtained powders were analyzed in order to determine their phase composition and thermal stability, morphology, dopants distribution, particles surface purity and luminescent characteristics. Owing to this, the chemical and crystal phase transformations that occurred  in specified periods of synthesis time are explained in detail. Moreover, it was shown that obtaining of uniformly doped (β) NaYF4:Yb,Er mosocrystalline particles is possible after 2h of sonication.

AB  - The Nonlinear Laser Scanning Microscopy (NLSM) contributes to the cell labeling through addressing two main issues: photobleaching and phototoxicity. Moreover, an increase of the penetration depth and a reduction of background autofluorescence are achieved.We have used a multidisciplinary approach combining expertise in material science, nanoparticles synthesis and characterization, cancer cell and tissue labeling, and high resolution imaging, in order to accomplish in vitro imaging of the cancer cells. We have imaged the oral squamous carcinoma cells and human gingival cells. We have demonstrated that we are able to take high contrast images. We have shown position of the nanoparticles in cells, through colocalization of the cell auto-fluorescence and the nanoparticles up-conversion.We plan to improve our abilities through further optimization of the up-converting nanoparticles (smaller and brighter particles) and microscopy technique.

abstract = "The Nonlinear Laser Scanning Microscopy (NLSM) contributes to the cell labeling through addressing two main issues: photobleaching and phototoxicity. Moreover, an increase of the penetration depth and a reduction of background autofluorescence are achieved.We have used a multidisciplinary approach combining expertise in material science, nanoparticles synthesis and characterization, cancer cell and tissue labeling, and high resolution imaging, in order to accomplish in vitro imaging of the cancer cells. We have imaged the oral squamous carcinoma cells and human gingival cells. We have demonstrated that we are able to take high contrast images. We have shown position of the nanoparticles in cells, through colocalization of the cell auto-fluorescence and the nanoparticles up-conversion.We plan to improve our abilities through further optimization of the up-converting nanoparticles (smaller and brighter particles) and microscopy technique.",

AB  - Monosized Up-Converting NanoParticles (UCNPs) with biocompatible surface and unique optical properties attract a great interest as new cell markers or drug delivery systems. The uppermost UC efficiency of β-NaYF4:Yb/Er phase is due to its hexagonal P63/m space group arrangement which could accommodate higher concentration of dopants at shorter distance. Stabilization of this phase in nanoparticles is usually achieved through thermal decomposition of organic precursors in the presence of solvents with a high boiling point. Here, for the same purpose, we used gadolinium co-doping during chitosan assisted solvothermal processing of inorganic precursor salts. Precursor concentration, solvent type, and synthesis time were varied in order to determine their influence on the β-NaY0.65Gd0.15F4:Yb0.18Er0.02 phase crystallization. The XRPD analysis showed that lower surplus of fluoride ions during synthesis leads to formation of Y0.65Gd0.15F4:Yb0.18Er0.02 orthorhombic phase, while the increase of fluoride content or prolongation of the processing time enhances formation α- NaY0.65Gd0.15F4:Yb0.18Er0.18 phase. Along with it, the changes of UCNPs morphology from spindle to spherical shape is detected. All samples emit intense green emission due to the (2H11/2, 4S3/2) → 4I15/2 electronic transitions, after been excited with infrared light (λ=978 nm).

With the increase in Gd3+ concentration, size of the nanoparticles decreased. The up-converting spectra recorded upon 980 nm irradiation showed the green (510-560 nm) and red (640-690 nm) emissions, due to 2H11/2, 4S3/2 → 4I15/2 and 4F9/2 → 4I15/2 transitions, respectively. The strongest up-conversion luminescence was detected in 15 mol% Gd3+-doped nanoparticles obtained after 20 hours of solvothermal treating. With the rise of Gd3+ content up-conversion emission decreased due to increased defect concentration in the NaYF4 matrix. Fourier transform infrared spectroscopy proved in situ generation of hydrophilic nanoparticles as a result of PVP ligands retention at the particle surface.

AB  - The synthesis of lanthanide doped up-converting nanoparticles (UCNPs), whose morphological, structural, and luminescence properties are well suited for applications in optoelectronics, forensics, security, or biomedicine, is of tremendous significance. The most commonly used synthesis method comprises decomposition of organometallic compounds in an oxygen-free environment and subsequent infliction of a biocompatible layer on the particle surface. In this work, hydroxyl-carboxyl (- OH/ - COOH) type of chelating agents (citric acid and sodium citrate) are used in situ for the solvothermal synthesis of hydrophilic NaY0.5Gd0.3F4:Yb,Er UCNPs from rare earth nitrate salts and different fluoride sources (NaF, NH4F, and NH4HF2). X-ray powder diffraction showed crystallization of cubic and hexagonal NaY0.5Gd0.3F4:Yb,Er phases in nano- and micro-sized particles, respectively. The content of the hexagonal phase prevails in the samples obtained when Na-citrate is used, while the size and shape of the synthesized mesocrystals are affected by the choice of fluoride source used for precipitation. All particles are functionalized with citrate ligands and emit intense green light at 519 nm and 539 nm (2H11/2, 4S3/2 → 4I15/2) under near infrared light. The intensity of this emission is distressed by the change in the origin of phonon energy of the host matrix revealed by the change in the number of the excitation photons absorbed per emitted photon.

AB  - The synthesis of lanthanide doped up-converting nanoparticles (UCNPs), whose morphological, structural, and luminescence properties are well suited for applications in optoelectronics, forensics, security, or biomedicine, is of tremendous significance. The most commonly used synthesis method comprises decomposition of organometallic compounds in an oxygen-free environment and subsequent infliction of a biocompatible layer on the particle surface. In this work, hydroxyl-carboxyl (- OH/ - COOH) type of chelating agents (citric acid and sodium citrate) are used in situ for the solvothermal synthesis of hydrophilic NaY0.5Gd0.3F4:Yb,Er UCNPs from rare earth nitrate salts and different fluoride sources (NaF, NH4F, and NH4HF2). X-ray powder diffraction showed crystallization of cubic and hexagonal NaY0.5Gd0.3F4:Yb,Er phases in nano- and micro-sized particles, respectively. The content of the hexagonal phase prevails in the samples obtained when Na-citrate is used, while the size and shape of the synthesized mesocrystals are affected by the choice of fluoride source used for precipitation. All particles are functionalized with citrate ligands and emit intense green light at 519 nm and 539 nm (2H11/2, 4S3/2 → 4I15/2) under near infrared light. The intensity of this emission is distressed by the change in the origin of phonon energy of the host matrix revealed by the change in the number of the excitation photons absorbed per emitted photon.

The formation of the hierarchically organized spherical aggregates and nanofoils was observed when CS and PAA were used during synthesis, respectively. The average crystallite size, microstrain, doping level, lattice parameters, as well as, the presence of the certain ligands on the particle surface were determined and correlated with the intensity of visible-light emission observed under 980 nm laser-diode excitation.

Due to efficient two-phonon excitation and the large anti-Stocks shift UCNPs are able to emit visible or UV photons under excitation by near-infrared (NIR). Over the last decade, decomposition of organometallic compounds has been indicated as one of the most convenient method for the synthesis of monodisperse NaYF4:Yb3+,Er3+ UCNPs with a hexagonal crystal structure. Herein, NaY0.8-xGdxYb0.18Er0.02F4 (x= 0.3 or 0.15) up-conversion nanoparticles crystallized in the hexagonal space group P63/m were successfully synthesized solvothermally utilizing rare earth nitrates, NaF and polyvinylpyrrolidone (PVP) in ethanol-water mixture at 200 °C. Rietveld refinement of the X-ray powder diffraction (XRPD) data and high resolution transmission microscopy (HRTEM) analysis show that all UCNPs are monocrystalline (60-70 nm), have low defect concentration and uniform dopants distribution. Fourier-transform infrared (FTIR) spectroscopy indicate existence of the PVP ligands at the UCNPs surface, while photoluminescence (PL) spectra shows characteristic green (at 520 and 540 nm, due to 2H11/2, 4S3/2→4I15/2 transitions) and red (at 655 nm, due to 4F9/2 → 4I15/2 transition) emission lines under excitation by NIR (λ =980 nm) light.

The x-ray powder diffraction (XRPD) showed that all powders contain the mixture of cubic and hexagonal NaYF4:Gd,Yb,Er phase in nano and micro-sized particles respectively. However, the content of later one prevails in samples obtain when Nacitrate is used as chelator, regardless of which fluoride source is used for precipitation. Additionally, variation of the particles size and shape is detected with a variation of fluoride type. All particles have hydrophilic surface due to retention of citrate ligands and emit intense green light emission centered at 519 and 539 nm (2H11/2,4S3/2→4I15/2) when excited with near infrared light.

AB  - Monosized up-converting nanoparticles (UCNPs) with a spherical shape and biocompatible surface have a wide application in biomedicine as new cell markers or light-triggered drug delivery agents. The synthesis of the hexagonal β-NaYF4:Yb/Er phase is of a great interest, because of its most efficient up-conversion luminescence. Beside it, synthesis of the UCNPs based on YF3:Yb/Er phase is also attractive due to its orthorhombic phase arrangement and fact that higher concentration of dopants could be introduced in such crystal lattice. In this work the synthesis of pure and Gd-doped YF3:Yb/Er phases were performed through hydro/solvo thermal method using a biocompatible chitosan as a surfactant. The XRD analysis showed that independently of the gadolinium content formation of the orthorhombic phase is achieved, but intensity of the green emission due to the (2H11/2, 4S3/2) → 4I15/2 electronic transitions was highest for un-doped YF3:Yb/Er sample.

AB  - Thanks to the unique optical properties, up-converting nanoparticles (UCNPs) have a wide application in optoelectronics, forensics, security and biomedicine. The synthesis of the most efficient hexagonal β-NaYF4: Yb/Er phase is usually performed through thermal decomposition of organic precursors which could cause the UCNP cytotoxicity. Since cubic polymorph is kinetically more stable than hexagonal, we used citric acid and Na-citrate for the nucleation of hexagonal NaYF4: Yb, Er phase in nanosized particles. Additionally, effect of different precipitation agents (NaF, NH4F and NH4HF2) used during solvothermal synthesis is explored. The XRPD analysis showed that using of citric acid led to a product composed from mixture of cubic and hexagonal NaYF4: Yb/Er phase, while the presence of Na-citrate influences the nucleation of well crystallized hexagonal β-NaYF4: Yb/Er phase, regardless of precipitation agents used. All samples are composed of polycrystalline spherical particles which size is influenced by the precursor chemistry. AB  - NaYF4 doped with Rare Earth elements such as Yb and Er is well known as a compound with the remarkable up-conversion photoluminesce and wide application. In this work, the influence of additional co-doping of NaYF4:Yb,Er with gadolinium (0, 15 and 30 mol%) was explored. The syntheses were performed solvothermally using the mixture of water/ethanol as medium and polyvinylpyrrolidone (PVP) as capping ligand. The obtained powders were subjected to X-ray powder diffraction (XRPD), scanning electron microscopy (SEM), Fourier-transform infrared (FTIR) spectroscopy and photoluminesce (PL) analyses. Structural refinement shown that NaYF4:Yb,Er particles generally crystallized in a cubic form (Fm-3m), while the additional presence of Gd3+ in the crystal lattice lead to the pure hexagonal phase crystallization (P63/m). Morphological analyses revealed that all powders are composed from nanodimensional particles with the size in the range of 50-70 nm, while FTIR spectra confirmed the presence of PVP functional groups on the particles surfaces. The strongest effect of Gd3+ doping was notices in the PL spectra, where the up-conversion response increases with the mol% of this dopant.

AB  - Lanthanide-doped fluoride up-converting nanoparticles (UCNPs) represent the new class of imaging contrast agents which hold great potential for overcoming existing problems associated with traditionally used dyes, proteins and quantum dots. In this study, a new kind of hybrid NaYF4:Yb,Er/PLGA nanoparticles for efficient biolabeling were prepared through one-pot solvothermal synthesis route. Morphological and structural characteristics of the as-designed particles were obtained using X-ray powder diffraction (XRPD), scanning and transmission electron microscopy (SEM/TEM), energy dispersive spectroscopy (EDS), Fourier transform infrared (FTIR) and photoluminescence (PL) spectroscopy, while their cytotoxicity as well as up-conversion (UC) labeling capability were tested in vitro toward human gingival cells (HGC) and oral squamous cell carcinoma (OSCC). The results revealed coexistence of the cubic (Fm-3m) and hexagonal (P63/m) phase in spherical and irregularly shaped nanoparticles, respectively. PLGA [Poly(lactic-co-glycolic acid)] ligands attached at the surface of UCNPs particles provide their enhanced cellular uptake and enable high-quality cells imaging through a near-infrared (NIR) laser scanning microscopy (λex = 980 nm). Moreover, the fact that NaYF4:Yb,Er/PLGA UCNPs show low cytotoxicity against HGC over the whole concentration range (10–50 μg/mL) while a dose dependent viability of OSCC is obtained indicates that these might be a promising candidates for targeted cancer cell therapy. © 2018 Elsevier B.V.

AB  - The emerging up-conversion nanoparticles (UCNPs) offer a wide range of biotechnology applications, from biomarkers and deep tissue imaging, to single molecule tracking and drug delivery. Their successful conjugation to biocompatible agents is crucial for specific molecules recognition and usually requires multiple steps which may lead to low reproducibility. Here, we report a simple and rapid one-step procedure for in situ synthesis of biocompatible amino-functionalized NaYF4:Yb,Er UCNPs that could be used for NIR-driven fluorescence cell labeling. X-ray diffraction showed that UCNPs synthesized through chitosan-assisted solvothermal processing are monophasic and crystallize in a cubic α phase. Scanning and transmission electron microscopy revealed that the obtained crystals are spherical in shape with a mean diameter of 120 nm. Photoluminescence spectra indicated weaker green (2H11/2, 4S3/2 → 4I15/2) and stronger red emission (4F9/2 → 4I15/2), as a result of enhanced non-radiative 4I11/2 → 4I13/2 Er3+ relaxation. The presence of chitosan groups at the surface of UCNPs was confirmed by Fourier transform infrared spectroscopy, thermogravimetry and X-ray photoelectron spectroscopy. This provides their enhanced internalization in cells, at low concentration of 10 μg ml-1, without suppression of cell viability after 24 h of exposure. Furthermore, upon 980 nm laser irradiation, the amino-functionalized NaYF4:Yb,Er UCNPs were successfully used in vitro for labeling of two human cell types, normal gingival and oral squamous cell carcinoma. © 2018 The Royal Society of Chemistry.

Polyacrilic Acid and Chitosan Assisted Solvothermal Synthesis of Up-converting NaYF4: Yb,Er Particles

AB  - There is a growing interest for development of a facile and reproducible approach  for the synthesis of biocompatible lanthanide doped up-converting nanoparticles (UCNPs) for deep tissue imaging and targeted drug delivery. Synthesis of such particles is usually performed through the decomposition of organometallic compounds, followed either with a ligands exchange or with a biocompatible layer coating. In this work, biocompatible NaYF4:Yb,Er (17 mol% Yb; 3 mol% Er) nanoparticles were synthesized by one-pot hydrothermal processing with an assistance of chitosan (Ch) or polyacrylic acid (PAA). Obtained powders were analyzed by X-ray powder diffraction (XRPD, Bruker D8 Discovery), field emission scanning electron microscopy (FE-SEM, Zeiss, DSM 960), transmission electron microscopy (TEM, JEOL JEM 2010), Fourier transform infrared (FTIR, Thermo Scientific Nicolet 6700) and photoluminescence (PL, Spex Fluorolog with C31034 cooled photomultiplier) spectroscopy. The results showed that although both powders crystallize in the same crystal arrangement (cubic, Fm-3m), particles size, shape and optical properties are dependent on the polymer used.

AB  - Lantanide dual-doped hydroxyapatite (HaP:Ln) is currently the subject of numerous studies in reconstructive medicine. Designed in form of hybrid nanoparticles which have magnetic and luminescent properties HaP:Ln (where Ln=Gd/Eu or Gd/Yb/Tm) is capable to enhance signal detection. Beside it, due biodegradable properties it is suitable for use in bone tissue engineering and target drug delivery. For such a promising approach, doping of a HAp matrix is performed with Gd/Eu and Gd/Yb/Tm during hydrothermal synthesis using EDTA as chelating agent. Morphological and structural characteristics of the particles were obtained using X-ray powder diffraction (XRPD), scanning and transmission electron microscopy (SEM/TEM), energy dispersive X-ray spectroscopy (EDX), Fourier Transform Infrared (FTIR) and photoluminescence (PL). The results show that needle-like nano- or micro- particles were obtained in all systems. Their phase composition and uniform distribution of dopants were confirmed by the structural refinement of the XRPD data and luminescence response from Eu and Tb (λex = 370, 394 and 977 nm).

AB  - In modern medical research, great attention has been focused to the development of the new biomarkers which include up-converting nanoparticles (UCNPs). Their optical response is triggered by NIR radiation that achieves deeper tissue penetration when compared with traditionally used fluorophores. In this work, biocompatible NaYF4: Yb, Er nanoparticles were synthesized by polymer assisted one-pot solvothermal processing using chitosan or poly(lacticco- glycolic acid). X-ray powder diffraction and electron microscopy results revealed differences in crystal arrangement and morphology of the as-synthesized particles. Fourier transform infrared spectroscopy confirmed the presence of corresponding polymers moiety on UCNPs surface providing their biocompatibility and low cytotoxicity towards human gingival fibroblasts (HFG). As a consequence of efficient up-conversion, prominent green emission (between 512-533nm and between 533-560nm) as well as red emission (630-690nm) were recorded in the particles photoluminescence spectra, and these are applied further in the visualization of the HFG using the laser scanning microscopy with a NIR laser source.
