AB  - Implantation of a biomaterial is one of the important trends in solving the problem of bone tissue loss. Calcium hydroxiapatite (HAp), as the most representative bone component is a serious candidate for such implantations. The synthetic polymer poly-L-lactide (PLLA) in HAp/PLLA is often used as a polymeric material, with a role in the substitution of bone tissue collagen fibers. Fibers of PLLA may strengthen HAp and its good bioresorption provides space for tissue remodeling. Differences in porosity, microstructure, compressive consistency as well as bioresorbility of HAp/PLLA may be achieved by using PLLA with different molecular weights. In this study HAp/PLLA composites with PLLA of different molecular weights (50,000; 160,000 and 430,000) were implanted in mouse peritoneum in order to examine the influence of the molecular weight of PLLA on morphology changes. Microstructural changes of biomaterial (HAp/PLLA) surface were analyzed one week, three weeks and four months after their implantation using Scanning Electron Microscopy. The results showed a significant difference in tissue reactions on the applied biocomposites, depending on their molecular weight. The most intense proliferation of cells was induced by HAp/PLLA 50,000 compared to HAp/PLLA 430,000 and HAp/PLLA 160,000. In the vicinity of HAp/PLLA 430,000 abundant erythrocytes were observed. The differences in biological reactions on the examined biocomposites are significant for their practical applications. HAp/PLLA composite biomaterials of different types and resorption rates require specific designing and programming to become suitable for particular purposes in an organism.

AB  - Početak XXI veka nesumnjivo je obeležen interdisciplinarnim i multidisciplinarnim naporima istraživača u različitim oblastima nauke. Jedna od najizrazitijih tendencija ovog tipa uočava se u biomedicinskim istraživanjima, gde se združuju napori lekara, biologa, genetičara i biohemičara, s jedne strane, i biofizičara i inženjera, s druge strane – sa ciljem dubljeg razumevanja zdravlja i bolesti, i primene ovih saznanja u biomedicinskoj praksi, tako važnoj u svakodnevnom životu ljudi.
Kao rezultat ovih svetskih trendova, u Srbiji već više godina na nekoliko fakulteta postoji nastava iz oblasti biomedicinskog inženjerstva, sa ciljem da osposobi inženjere ovih usmerenja za multidisciplinarno povezivanje znanja iz oblasti tehnike sa biomedicinskim znanjima. Jedan od bazičnih predmeta ovih usmerenja jesu Biomaterijali, kojima je i posvećen naš udžbenik, čiji je cilj da predstavi pregled teorije i prakse biomaterijala u biomedicinskoj nauci.
Nauka o biomaterijalima je nesumnjivo najmultidisciplinarnija od svih nauka, jer zahteva ovladavanje znanjima iz mnogih oblasti nauke i tehnologije, inženjerstva i medicine, kako bi naučnici iz oblasti biomaterijala mogli da se uhvate u koštac sa ovom profesijom. Zato posle uvodnog dela, udžbenik iz Biomaterijala sadrži četiri celine: (I) Osnovni biomedicinski koncepti i reakcije organizma na biomaterijale, (II) Struktura, fizičko-mehanička karakterizacija i modeliranje biomaterijala i tkiva, (III) Savremeni biomaterijali i tehnologije, (IV) Perspektive biomaterijala i tehnologija, iza kojih slede Zadaci sa rešenjima, Ispitna test pitanja i Ispitna teorijska pitanja, koji pomažu studentima da lakše savladaju veoma obimno i kompleksno gradivo. Na kraju svakog poglavlja data su pitanja za rekapitulaciju, kao i spisak dopunske literature za opcionu detaljniju obradu pojedinih oblasti.
Grupa od dvadeset četiri profesionalca sa univerziteta i naučnih instituta, pod okriljem Instituta tehničkih nauka Srpske akademije nauka i umetnosti, Beograd, i Društva za istraživanje materijala Srbije (MRS Srbija) doprinela je pisanju ovog kapitalnog udžbenika o biomaterijalima, prvog do sada na srpskom jeziku. Mada uključivanje veće grupe autora nužno dovodi do stilske neujednačenosti, ipak je oblast biomaterijala toliko multidisciplinarna da je ovakav pristup bio neophodan, kako uostalom pokazuju slična svetska iskustva sa uključivanjem i preko pedeset autora. Ipak urednici su se potrudili da koliko je to moguće stilski i pedagoški ujednače udžbenik, kako bi bio korisna literatura za sve studente diplomskih, master i doktorskih studija iz biomedicinskog inženjerstva u Srbiji i okruženju.

Changes of HAp/PLLA biocomposites and tissue reaction after subcutaneous implantation

AB  - Experimental and clinical studies have shown that HAp granules and powder may be successfully applied in reconstruction of bone defects. Composite HAp/PLLA biomaterial embodies good characteristics of each of these biomaterials. Differences in porosity, microstructure, compressive consistency as well as in bioresorbility of HAp/PLLA are achieved by using PLLA of different mole masses. The aim of this study was to analyse light microscopy of the tissue reaction and changes of implants made of combination of syngeneic bone fragments and HAp/PLLA biocomposites with PLLA molecule mass of 430 000 (HP2) and 50 000 (HP3). The mice were divided into 4 experimental studies, 8 animals each. Two groups underwent implantation one group of mice was falsely operated, and one group was used for general control. Pathohistological analysis of implants was performed three months after subcutaneous implantation. Phagocytosis of HAp/ PLLA and bone was noticed on both implants’ preparations. Blood vessels were also noticed confirming the process of angiogenesis around the implants’ particles. The holes around particles of biocomposites and bone fragments represented the signs of their resorption. The presence of multinuclear cells between implants’ particles was observed and it was more prominent in HP3 implants. Osteogenesis is more intense in HP3 and ectopic hematopoesis is present. Our results show that designed HAp/PLLA biocomposites have good biocompatibility, their combination with bone fragments of a vital bone enables osteogenesis and hematopoesis and these processes are more prominent if PLLA biocomposite with lower molecular mass was used.

AB  - Chemically synthesized hydroxyapatite/poly-l-lactide (HAp/PLLA) composite biomaterial was studied in vivo. The biocomposite was implanted into Balb/c Singen mice and after 1 and 3 weeks removed from their organisms and analyzed by the FT-IR spectroscopy. After 1 week of testing in vivo the implanted sample gave a spectrum in which absorption bands arising from newly formed functional groups of amine and peptide can be seen. After 3 weeks, a spectrum with pronounced absorption bands at 3420 and 1650 cm−1 assigned to newly generated collagen, a component of the extracellular connective-tissue matrix, was registered. Also, decrease of the intensity absorption band at 1760 cm−1 originating from the CO group of PLLA indicates bioresorption of the PLLA used. Analysis of the microstructure of the sample surface by scanning electron microscopy before and after implantation revealed bioresorption of the PLLA polymer phase and generation of collagen fibers at the sites of implanted bioresorptive PLLA. A mixture of autologous bone powder and HAp/PLLA biocomposite was also examined. After implantation, the same final products as in the case of HAp/PLLA composite biomaterial used alone were found.

After 1 week of testing in vivo the implanted sample gave a spectrum in which absorption bands arising from newly formed functional groups of amine and peptide can be seen. After 3 weeks, a spectrum with pronounced absorption bands at 3420 and 1650 cm−1 assigned to newly generated collagen, a component of the extracellular connective-tissue matrix, was registered. Also, decrease of the intensity absorption band at 1760 cm−1 originating from the CO group of PLLA indicates bioresorption of the PLLA used. Analysis of the microstructure of the sample surface by scanning electron microscopy before and after implantation revealed bioresorption of the PLLA polymer phase and generation of collagen fibers at the sites of implanted bioresorptive PLLA. A mixture of autologous bone powder and HAp/PLLA biocomposite was also examined. After implantation, the same final products as in the case of HAp/PLLA composite biomaterial used alone were found.",

AB  - Besides its high osteoinductive properties, hydroxyapatite (HAp) exhibits a relatively low mechanical strength. In order to improve the mechanical properties and reliability of HAp based composites, the addition of selected polymers is highly recommended, The main objective of this work is to study the microstructural characteristics of HAp/poly-L-lactide (PLLA) composites obtained by cold or hot processing. The composites were prepared from a mixture of a chloroform solution of poly-L-lactide with granulated HAp. After elimination of chloroform by vacuum evaporation, dense compacts were obtained by cold or hot pressing. The pressing pressure ranged from 98.10 to 294.3 MPa for both cold and hot pressing, The hot pressing was performed in the temperature region 293-457 K for a time period of 15-60 min. Depending on the PLLA amount and the pressing procedure it is possible to obtain highly porous or nearly fully dense composites, The scanning electron microscopy examination of fracture as well as of free surfaces revealed that the final porosity and wetting are affected to a great extent by the synthesis conditions and amount of polymer added. An increase in temperature to 457 K for a longer period of time results in fully dense compacts, The formation of a nearly continuous polymer network that leads to the hardening of HAp has also been observed. However, it should be pointed out that some layers of HAp may be free of polymer film since PLLA penetrates more deeply into the porous HAp.

AB  - Calcium hydroxyapatite (HAp) and poly-L-lactide (PLLA) were synthesized chemically. The obtained HAp was of high purity and, after special thermal treatment, of high crystallinity as well. Synthesis of PLLA was performed using L-lactide as a monomer and nontoxic initiator. In this way a polymer of large molar weight (about 400 000) was obtained. The HAp and PLLA obtained were used as constituents of the HAp/PLLA composite biomaterial, a potential material for implants. The composite was obtained by mixing completely dissolved PLLA with granules of HAp. The composite was compacted by cold and hot pressing at pressures of 49.0-490.5 MPa and temperatures of 20-184 degrees C. The material obtained at optimum process parameters had a density of 99.6% and compressive strength of 93.2 MPa.
