AB - Immobilizing antifungal polyenes such as nystatin (Nys) and amphotericin B (AmB) into biodegradable formulations is advantageous compared to free drug administration providing sustained release, reduced dosing due to localized targeting and overall reduced systemic drug toxicity. In this study, we encapsulated Nys and AmB in medium chain length polyhydroxyalkanoates (mcl-PHA) microspheres (7–8 µm in diameter). The obtained formulations have been validated for antifungal activity in vitro against a panel of pathogenic fungi including species of Candida, Aspergillus, Microsporum and Trichophyton genera and toxicity and efficacy in vivo using the zebrafish model of disseminated candidiasis. While free polyenes, especially AmB, were highly toxic to zebrafish embryos at the effective (MIC) doses, after their loading into mcl-PHA microspheres, inner organ toxicity and teratogenicity associated with both drugs were not observed, even at 100 × MIC doses. The obtained mcl-PHA/polyene formulations have successfully eradicated C. albicans infection and showed an improved therapeutic profile in zebrafish by enhancing infected embryos survival. This approach is contributing to the antifungal arsenal as polyenes, although the first broad-spectrum antifungals on the market are still the gold standard for treatment of fungal infections.

AB - We report an innovative low-cost wet precipitation synthesis method for gelatin-modified zinc oxide nanoparticles (GM ZnO NPs) at the inter- face between the gelatin hydrogel and aqueous elec- trolyte. Diffusion of ammonia through the hydrogel matrices with different gelatin contents induced pre- cipitation of the product in contact with the surface of the aqueous solution of zinc ions. The obtained precipitate was subjected to thermal treatment to partially decompose the adsorbed gelatin in the NP structure. Physicochemical properties of obtained GM ZnO NPs were characterized by X-ray powder diffraction (XRD), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), differential thermal analysis (DTA), thermo- gravimetry (TG), photon correlation spectroscopy (PCS), zeta potential measurements, and inductively coupled plasma-mass spectrometry (ICP-MS). The estimated mean crystallite size of GM ZnO NP pow- ders was in the range from 5.8 to 12.1 nm. The syn- thesized NPs exhibited nanosheet morphology and arranged into flake-like aggregates. The toxic poten- tial was investigated in vitro in human hepatocellular carcinoma cell line HepG2. The thiazolyl blue tetra- zolium bromide (MTS) assay was used to assess cell viability, 2′,7′-dichlor-fluorescein-diacetate (DCFH- DA) assay to examine the formation of intracellu- lar reactive oxygen species (ROS), and comet assay to evaluate the genotoxic response. GM ZnO NPs slightly reduced HepG2 cell viability, did not induce ROS formation, and showed low genotoxic potential at very high doses (100 μg mL−1). ZnO NPs fabri- cated and modified using the proposed methodol- ogy deserve further study as potential candidates for antibacterial agents or dietary supplements with low overall toxicity.

AB - The catalytic activity of the walnut shell ash was investigated in the biodiesel production by the sunflower oil methanolysis. The catalyst was characterized by the TG-DTA, XRD, Hg porosimetry, N2 physisorption, SEM, and Hammett method. In addition, the effects of the catalyst loading and the methanol-to-oil molar ratio on the methyl esters synthesis were tested at the reaction temperature of 60 °C. The walnut shell ash provided a very fast reaction and a high FAME content (over 98%). As the reaction occurred in the absence of triacylglycerols mass transfer limitation, the pseudo-first-order model was employed for describing the kinetics of the reaction. The catalyst was successfully reused four times after the regeneration of the catalytic activity by recalcination at 800 °C.

AB - Composite hydrogels capable of controlled drug delivery via ion exchange are an interesting group of materials for the construction of implantable drug reservoirs for electrically charged drugs. In this study, we synthesized composite poly(DL-lactide-co-glycolide)/poly(acrylic acid) (PLGA-PAA) hydrogels by sequential application of UV or gamma irradiation and traditional phase inversion. Physicochemical properties of the composite PLGA-PAA hydrogels were investigated using Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR), scanning electron microscopy (SEM), and differential scanning calorimetry (DSC). We examined the ion exchange capacity (IEC) and swelling behavior of these materials to determine their potential as drug reservoirs. Composite PLGA-PAA hydrogel synthesized using UV irradiation (UV-PLGA-PAA) exhibited a porous microstructure with submicron-sized hydrogel-rich aggregates and homogeneous chemical composition. Swelling behavior and IEC of this material were highly reproducible. Composite PLGA-PAA hydrogels synthesized using gamma irradiation (G-PLGA-PAAs) had a less uniform microstructure with larger pores and micron-sized hydrogel-rich aggregates while exhibiting rather inhomogeneous chemical composition. These materials showed superior swelling properties, but a more variable IEC, compared to the material fabricated using UV irradiation. Results of DSC analysis showed a dose-dependent decrease in glass transition temperature for G-PLGA-PAAs indicating the effects of PLGA chain scission. Our findings indicate that gamma irradiation is a possible alternative to UV irradiation in the synthesis of composite PLGA-PAA hydrogels which can modify or control important material properties. However, the synthesis protocol using gamma irradiation should be further optimized to improve the IEC reproducibility. In our future research, we will investigate the in vitro release of charged drugs from synthesized composite PLGA-PAA hydrogels under physiological conditions.

AB - The aim of this study was to improve the mechanical properties and to optimize antimicrobial activity of hydroxyapatite (HAP) by simultaneous doping with Mg and Cu ions in order to obtain material that would be able to assist in the bone/tooth healing process, prevent post-implementation infections and provide satisfying values of hardness and fracture toughness for biomedical application. Ion doping was done during the hydrothermal synthesis of HAP powders, whereby the content of Mg ions in the starting solution was varied between 1-20 mol. % with regard to Ca ions, while the amount of Cu ions was kept constant at 0.4 mol. %. The green compacts were sintered for 2 h at temperatures ranging 750–1200 °C depending on the Mg content, chosen in agreement with dilatometry results. Presence of Mg ions was found to favour transition from HAP to β−tricalcium phosphate phase (β−TCP), which enabled formation of biphasic HAP/β−TCP and pure β−TCP phase at 160 °C during hydrothermal synthesis. In vitro investigation of antimicrobial activity against Escherichia coli, Staphylococcus aureus and Enterococcus faecalis showed satisfactory antimicrobial activity. MTT assay performed on MRC-5 and L929 cell lines showed excellent cytocompatibility and cell proliferation. Maximum hardness by Vickers and fracture toughness values, 4.96 GPa and 1.75 MPa m1/2 respectively, were obtained upon addition of 5 mol. % Mg, as a consequence of the lowest grain size and porosity, as well as the highest densification rate. This is, to the best of our knowledge, the highest fracture toughness for HAP or β-TCP ceramics reported thus far.

AB - Nanostructured zinc oxides are promising materials for numerous biomedical applications where they can serve as therapeutic agents or tools for sensing and imaging. Despite their favorable properties, wider use of zinc oxide nanoparticles in biomedicine is limited by toxicity issues. Therefore, new synthesis approaches should be devised to obtain zinc oxide nanoparticles which are safe-by-design. We present an innovative low-cost wet precipitation synthesis of gelatin modified zinc oxide nanoparticles at the gel/liquid interface. The diffusion of ammonia through the gelatin hydrogels of different porosities induces precipitation of the product in contact with the surface of the aqueous solution of zinc ions. After thermal treatment of the precipitate, adsorbed organic residues of decomposed gelatin act as modifiers of zinc oxide nanoparticles. We characterized the physicochemical properties of obtained gelatin modified zinc oxide nanoparticles by XRD, FTIR, DTA/TG, and SEM. The synthesized nanoparticles show hexagonal wurtzite structure and form flakelike aggregates. FTIR and DTA/TG analyses indicate that the thermal decomposition of adsorbed gelatin depends on the gelatin content of the hydrogel used in the synthesis. We also examined the viability of HepG2 cells, generation of intracellular reactive oxygen species, and genotoxicity using the MTS, DCFH-DA, and alkaline comet assay, respectively. Fabricated gelatin modified zinc oxide nanoparticles show very low toxicity potential at doses relevant for human exposure.

AB - Bioactive glasses have been used for over three decades in biomedical applications owing to high bioactivity, biocompatibility, as well as the possibility to stimulate regeneration of the bone. The aim of this work was to synthesized bioactive glasses, which contain lithium and strontium, by commercial method melting-quenching, as well as determining the properties, affected by mentioned ions. Differential thermal/thermogravimetric analysis, particle size distribution, energy-dispersive X-ray spectroscopy, inductively coupled plasma optical emission spectrometry, evaluation of the antimicrobial activity, in vitro bioactivity and biocompatibility test and scanning electron microscopy were used for characterization. The results showed that glass transition and crystallization temperatures are decreasing with addition of lithium and strontium ions. Furthermore, the samples showed high inhibition rate of Escherichia coli growth, as well as high bioactivity and biocompatibility. The crystal apatite layer, formed on the surface of synthesized glasses after seven days in simulated body fluid, differs in shape, size and interconnection of the glass particles, which depends on concentration of lithium and strontium ions.

AB - The densification behavior of hydroxyapatite nanorods prepared by chemical precipitation method in open reactor conditions was investigated by application of different heating rates. The non-isothermal processing was performed with 2, 10, and 50 °C/min up to 1200 °C and yielded fully dense ceramics. The implementation of the higher heating rate provided grain size refinement from micrometer level for the slowest ramp, down to 250 nm in the case of processing with 50 °C/min, without any drawbacks regarding final density. The relative amount of retained structural hydroxyl groups in sintered ceramics was gradually increased with the heating rate. Furthermore, the qualitative level of optical translucency was increased with a higher heating rate which can be explained by the beneficial alignment of HAp nanorods during the fast heating rate processing, achieved microstructural refinement, and higher amount of structural hydroxyl groups.
