AB  - The aim of this work is in situ anodization/anaphoretic deposition of a nano amorphous calcium phosphate (ACP)/chitosan oligosaccharide lactate (ChOL) multifunctional hybrid coating decorated with selenium (Se) on a titanium substrate and in vivo investigation of its immunomodulatory and anti-inflammatory effect. Investigating phenomena at the implant–tissue interface of interest for controlled inflammation and immunomodulation was also the aim of the research. In our earlier research, we designed coatings based on ACP and ChOL on titanium with anticorrosive, antibacterial and biocompatible properties, while in the presented results we show that selenium addition makes this coating an immunomodulator. The immunomodulatory effect of the novel hybrid coating is characterized by the examination of the functional aspects in the tissue around the implant (in vivo): proinflammatory cytokines’ gene expression, M1 (iNOS) and M2 (Arg1) macrophages, fibrous capsule formation (TGF-β) and vascularization (VEGF). The EDS, FTIR and XRD analyses prove the formation of a ACP/ChOL/Se multifunctional hybrid coating on Ti and the presence of Se. A higher M2/M1 macrophage ratio in the ACP/ChOL/Se-coated implants compared to pure titanium implants (a higher level of Arg1 expression) is noted at all time points examined (after 7, 14 and 28 days). Lower inflammation measured by gene expression of proinflammatory cytokines IL-1β and TNF, lower expression of TGF-β in the surrounding tissue and higher IL-6 expression (solely at day 7 post-implantation) is noted in presence of the ACP/ChOL/Se-coated implants.

AB  - Collagen membranes are routinely used in oral surgery for bone regeneration. Despite their numerous advantages, such as stimulating bone growth, bacterial contamination still remains one of the disadvantages of membrane use. Thus, we assessed the biocompatibility and osteogenic and antibacterial properties of a collagen membrane (OsteoBiol) modified with chitosan (CHI) and hydroxyapatite nanoparticles (HApNPs). Attenuated total reflectance-Fourier transform infrared spectroscopy (ATR FT-IR), X-ray powder diffraction (XRD), and field emission scanning electron microscopy (FE-SEM) were performed for membrane characterization. Biocompatibility was assessed on dental pulp stem cells (DPSCs) by an MTT assay, while the osteogenic effect was assessed by an ALP activity assay and qPCR analysis of osteogenic markers (BMP4, ALP, RUNX2, and OCN). Antimicrobial properties were investigated by counting colony-forming units (CFUs) of Streptococcus mitis, Porphyromonas gingivalis, and Fusobaterium nucleatum on membranes and in the surrounding medium. Membranes showed no cytotoxicity. ALP activity was higher and ALP, BMP4, and OCN genes were up-regulated in DPSCs on modified membranes compared to unmodified membranes. The CFUs were reduced on modified membranes and in the medium. Modified membranes showed great biocompatibility and a high osteoinductive effect. Additionally, they showed antimicrobial and antibiofilm effects against periopathogens. It can be concluded that the incorporation of CHI and hydroxyapatite nanoparticles in collagen membranes may be advantageous to promote osteogenesis and reduce bacterial adhesion.

AB  - Hydroxyapatite attracts great attention as hard tissues implant material for bones and teeth. Its application in reconstructive medicine depends on its biocompatibility, which is in a function of composition and surface properties. The insertion of a protein element in the composition of implants can improve the cell adhesion and the osseointegration. Having this in mind, the proposal of this work was to develop L-alanine-grafted hydroxyapatite nanoparticles and to study their biocompatibility. Two L-alanine sources and three grafting methods were used for hydroxyapatite surface functionalization. The efficiency of grafting was determined based on X-ray powder diffraction, Fourier-transform infrared spectroscopy, thermal analyses, and field-emission scanning electron microscopy. The results indicated the formation of hydroxyapatite with 8–25 wt% of organic content, depending on the grafting method. Protein adsorption, cell adhesion, and viability studies were carried out to evaluate biological properties of grafted materials. The viability of MG-63 human osteoblastic cells following 24 h incubation with the alanine-grafted hydroxyapatite samples is well preserved, being in all cases above the viability of cells incubated with hydroxyapatite. The alanine-grafted hydroxyapatite prepared in situ and by simple mixture showed higher protein adsorption and cell adhesion, respectively, indicating their potential toward use in regenerative medicine.

Effect of argon plasma pre-treatment of healing abutments on peri-implant microbiome and soft tissue integration: a proof-of-concept randomized study

AB  - Purpose: Biofilm-free implant surface is ultimate prerequisite for successful soft and bone tissue integration. Objective of the study was to estimate the effects of argon plasma healing abutment pre-treatment (PT) on peri-implant soft-tissue phenotype (PiSP), inflammation, plaque accumulation and the microbiome (PiM) between non-treated (NPT) and treated (PT) abutments following 3-months healing period. The hypothesis was that cell-conductive and antimicrobial properties of PT would yield optimal conditions for soft tissue integration.
Material and Methods: Two months following second-phase surgery, microbiological and clinical parameters were assessed around thirty-six healing abutments with two types of microtopography, smooth surface (MACHINED) and ultrathin threaded microsurface (ROUGH). A two level randomization schema was used to achieve equal distribution and abutments were randomly divided into rough and machined groups, and then divided into PT and NPT groups. PiM was assessed using next-generation DNA sequencing.
Results: PiM bacterial composition was highly diverse already two months post-implantation, consisting of key-stone pathogens, early and late colonizers, while the mycobiome was less diverse. PT was associated with lower plaque accumulation and inflammation without significant impact on PiSP, while in NPT clinical parameters were increased and associated with periopathogens. NPT mostly harbored late colonizers, while PT exerted higher abundance of early colonizers suggesting less advanced plaque formation. Interaction analysis in PT demonstrated S. mitis co-occurrence with pro-healthy Rothia dentocariosa and co-exclusion with Parvimonas micra, Porphyromonas endodontalis and Prevotella oris. PiSP parameters were generally similar between the groups, but significant association between PiM and keratinized mucosa width was observed in both groups, with remarkably more expressed diversity in NPT compared to PT. PT resulted in significantly lower BOP and PI around rough and machined abutments, respectively, without specific effect on PiM and PiSP.
Conclusions: PT contributed to significantly the less advanced biofilm accumulation and inflammation without specific effects on PiSP.

abstract = "Purpose: Biofilm-free implant surface is ultimate prerequisite for successful soft and bone tissue integration. Objective of the study was to estimate the effects of argon plasma healing abutment pre-treatment (PT) on peri-implant soft-tissue phenotype (PiSP), inflammation, plaque accumulation and the microbiome (PiM) between non-treated (NPT) and treated (PT) abutments following 3-months healing period. The hypothesis was that cell-conductive and antimicrobial properties of PT would yield optimal conditions for soft tissue integration.
Material and Methods: Two months following second-phase surgery, microbiological and clinical parameters were assessed around thirty-six healing abutments with two types of microtopography, smooth surface (MACHINED) and ultrathin threaded microsurface (ROUGH). A two level randomization schema was used to achieve equal distribution and abutments were randomly divided into rough and machined groups, and then divided into PT and NPT groups. PiM was assessed using next-generation DNA sequencing.
Results: PiM bacterial composition was highly diverse already two months post-implantation, consisting of key-stone pathogens, early and late colonizers, while the mycobiome was less diverse. PT was associated with lower plaque accumulation and inflammation without significant impact on PiSP, while in NPT clinical parameters were increased and associated with periopathogens. NPT mostly harbored late colonizers, while PT exerted higher abundance of early colonizers suggesting less advanced plaque formation. Interaction analysis in PT demonstrated S. mitis co-occurrence with pro-healthy Rothia dentocariosa and co-exclusion with Parvimonas micra, Porphyromonas endodontalis and Prevotella oris. PiSP parameters were generally similar between the groups, but significant association between PiM and keratinized mucosa width was observed in both groups, with remarkably more expressed diversity in NPT compared to PT. PT resulted in significantly lower BOP and PI around rough and machined abutments, respectively, without specific effect on PiM and PiSP.
Conclusions: PT contributed to significantly the less advanced biofilm accumulation and inflammation without specific effects on PiSP.",

AB  - Sn-Pd electrocatalysts with a constant atomic ratio of 60 at.% Sn‒40 at.% Pd suitable for potential application in direct ethanol fuel cells were synthesized using a novel two-step electrodeposition method. First, Sn was electrodeposited in various forms of dendrites, from spear-like and needle-like to individual fern-like dendrites to a network of intertwined fern-like dendrites, by varying the cathodic potential and then performing electrodeposition of Pd at a constant current density in the second step. A morphological and elemental analysis of Sn and Sn-Pd electrocatalysts was performed using scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) techniques, while the size of Sn dendrites was analyzed using the particle size distribution (PSD) method. Cyclic voltammetry (CV) and chronoamperometry were applied in order to study the catalytic behavior of Sn-Pd electrocatalysts in the ethanol oxidation reaction (EOR), while CO stripping was used to estimate the antipoisoning capability of the electrocatalysts. The Sn surface morphology of the sub-layer was highly correlated with the electrocatalytic activity of the examined Sn-Pd electrocatalysts. The high activity it presented towards the EOR showed the suitability of the Sn-Pd electrocatalyst constructed from individual fern-like Sn dendrites as a sub-layer. Compared to Pd alone, this Sn-Pd catalyst showed more than 3 times higher activity and improved EOR kinetics. This enhancement in the catalytic activity of the Sn-Pd electrocatalysts is attributed to both the morphological characteristics of Sn as a sub-layer and the bifunctional effect.

AB  - Dense calcium phosphate-based ceramics were fabricated to be used as targets for pulsed laser deposition (PLD). Nanostructured cobalt-substituted hydroxyapatite (Co:HAP) was used as a starting powder. To vary phase composition and microstructure of targets, two sintering approaches were applied, conventional (CS) and two-step sintering (TSS). The obtained results show that in both cases biphasic calcium phosphate (BCP) ceramics (targets) were prepared, with slightly different HAP-to-β–TCP amount ratio and a significantly different microstructure. While the CS method yielded fully dense ceramics with an average grain size of 1.3 μm, the ceramics prepared by TSS had a density of 98.5%, with a predominant grain size below 100 nm. (Ca + Co)P coatings were prepared by PLD of (Ca + Co)P targets. The temperature of the Ti substrate was adjusted to be 25 and 500 °C. The results show that the phase composition of (Ca + Co)P coatings depended on the phase composition of targets as well as on the temperature of the Ti substrate. The coating prepared at 25 °C using CS target consisted of three calcium phosphate phases, HAP, β–TCP and α–TCP; when the TSS target was used, the coating was biphasic, containing HAP and β–TCP. When the substrate was heated to 500 °C, regardless of whether the CS or the TSS target was used, the deposited coatings were composed of HAP and α–TCP. Due to different phase compositions, the (Ca + Co)P coatings deposited at 25 °C showed an improved hardness compared to those deposited at 500 °C. The obtained results confirmed that the phase composition, morphology and mechanical properties of 0.3 μm thick (Ca + Co)P coatings on a Ti substrate can be tailored by employing (Ca + Co)P targets with different microstructures, and also by varying the temperature of the Ti substrate during deposition experiments.

(Ca + Co)P coatings were prepared by PLD of (Ca + Co)P targets. The temperature of the Ti substrate was adjusted to be 25 and 500 °C. The results show that the phase composition of (Ca + Co)P coatings depended on the phase composition of targets as well as on the temperature of the Ti substrate. The coating prepared at 25 °C using CS target consisted of three calcium phosphate phases, HAP, β–TCP and α–TCP; when the TSS target was used, the coating was biphasic, containing HAP and β–TCP. When the substrate was heated to 500 °C, regardless of whether the CS or the TSS target was used, the deposited coatings were composed of HAP and α–TCP. Due to different phase compositions, the (Ca + Co)P coatings deposited at 25 °C showed an improved hardness compared to those deposited at 500 °C. The obtained results confirmed that the phase composition, morphology and mechanical properties of 0.3 μm thick (Ca + Co)P coatings on a Ti substrate can be tailored by employing (Ca + Co)P targets with different microstructures, and also by varying the temperature of the Ti substrate during deposition experiments.

AB  - Hydroxyapatite (HAp) attracts great attention due to application in reconstructive medicine for hard tissues, mostly bones and teeth, where it is declared to be highly biocompatible material. Its grafting with amino acids further increases biocompatibility and has crucial importance for acceptance of body implants. In this work different methods of grafting were investigated: simple mixing, thermal treatment induction and in situ synthesis/grafting reactions. Two amino acid precursors were separately tested in grafting procedures: pure alanine and alanine methyl ester hydrochloride. The efficiency of grafting was determined based on X-ray powder diffraction (XRPD), Fourier-transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and thermal analyses (DSC, TG/DTA) of obtained powders, while complementary UV-VIS spectroscopy of supernatants was additionally performed for quantitative determination of non-grafted nitrogen using ninhydrin standardized procedure.

AB  - This paper deals with the deposition of calcium phosphate and hybrid coatings based on calcium phosphate ceramics on titanium with improved properties by in situ anodization/anaphoretic deposition process. Hybrid coatings consisted of chitosan oligolactate (ChOL) and ChOL with Se as immunomodulatory oligoelement. The paper contributes to solving the problem of multi-stage pre-treatment and post-treatment of titanium and oxidized titanium surface to obtain a coating on the substrate, adhesion of the coating, antimicrobial and cytotoxic properties that occur in biomaterials, as well as reduced immune inflammatory response of the organism. The in situ method results in improved adhesion bioactive coating, increased bioactivity and biocompatibility with increased antimicrobial properties and absence of cytotoxicity. The studied biomaterials have improved properties such as: corrosion resistance, absence of toxicity to the human body and adequate strength, which enables their potential use in medicine and dentistry. The innovation is reflected in combining calcium phosphate coating synthesis and surface modification by partially incorporating a ceramic coating into the crystalline structure of the substrate. The characterization of the coatings obtained in this manner was performed by various physico-chemical, biochemical and biological methods. These characterization techniques included: FE-SEM, XRD, FTIR, cytotoxicity and in vivo testing.

It primarily deals with the creation and optimization of a new in situ anodizing/anaphoretic deposition process for obtaining multifunctional composite biomaterials. The in situ method results in improved adhesion bioactive coating, increased bioactivity and biocompatibility with increased antimicrobial properties and absence of cytotoxicity. The studied biomaterials have improved properties such as: corrosion resistance, absence of toxicity to the human body and adequate strength, which enables their potential use in medicine and dentistry. AB  - Тестирање биоматеријала намењених за примену у регенеративној медицини на ћелијским моделима in vitro је неопходан први корак у циљу процене биокомпатибилности биоматеријала али и њихове биофункционалности. Постоје различити начини тестирања биоматеријала, као и различити ћелијски модели на којима се врши тестирање. Одабир адекватног ћелијског модела врши се на основу врсте биоматеријала и његове потенцијалне примене а тестирање се врши најпре стандардним тестовима као што су тестови цитотоксичности и генотоксичности а затим се испитују друге биолошке карактеристике као што су нпр. способност биоматеријала да индукује диференцијацију ћелија у одређени тип или способност активације макрофага ка одређеном фенотипу и индукција њиховог одговора ин витро, и многе друге. У овом раду ће бити преглед резултата наших досадашњих истраживања у области карактеризације биоматеријала и испитивања на ћелијским моделима in vitro. Испитивали смо биоматеријале намењене за регенерацију тврдих ткива, нпр. материјали на бази хидроксиапатита, калцијум фосфата, природног и синтетског порекла, тако и биоматеријале за регенерацију меких ткива, нпр. материјали на бази колагена или других полимера како природног тако и синтетског порекла. Да би се побољшала биофункционалност биоматеријала често им се додају биоактивне супстанце или њихове комбинације, а материјали могу бити у различитим облицима, у виду скафолда, у виду гранула, нано- или микро- димензија, мембрана и разним другим, при чему облик материјала утиче и на одабир ћелијског модела који се користи за тестирање. За карактеризацију биматеријала користили смо методе као што су SEM, EDX, FTIR и друге. Рађени су и тестови цитотоксичности, генотоксичности, пролиферације, миграције, диференцијације, фагоцитозе и многи други, а биоматеријале смо тестирали у директном контакту са ћелијама и у индиректном контакту. Испитивали смо и промене биоматеријала након интеракције са ћелијама као и понашање биоматеријала у различитим условима in vitro.
AB  - Testing biomaterials intended for use in regenerative medicine on cell models in vitro is a necessary first step in order to assess the biocompatibility of biomaterials as well as their biofunctionality. There are different methods of testing biomaterials, as well as different cell models on which testing is performed. The selection of an adequate cell model is made based on the type of biomaterial and its potential application, and testing is performed first using standard tests such as cytotoxicity and genotoxicity tests, and then other biological characteristics are examined such as, for example, the ability of biomaterials to induce the differentiation of cells into a certain type or the ability to activate macrophages towards a certain phenotype and the induction of their response in vitro, and many others. This paper will present the results of our research in the field of characterization of biomaterials and examination on cell models in vitro. We examined biomaterials intended for the regeneration of hard tissues, e.g. materials based on hydroxyapatite, calcium phosphate, of natural and synthetic origin, as well as biomaterials for the regeneration of soft tissues, e.g. materials based on collagen or other polymers of both natural and synthetic origin. In order to improve the biofunctionality of biomaterials, bioactive substances or their combinations are often added, and the biomaterials can be in different forms, in the form of scaffolds, in the form of granules, nano- or micro-dimensions, membranes and many others, whereby the shape of the material also affects the selection of the cell model used for testing. To characterize bimaterials, we used methods such as SEM, EDX, FTIR and others. In addition to cytotoxicity and genotoxicity tests, we performed proliferation, migration, differentiation, phagocytosis and many other tests, and we tested biomaterials both in direct contact with cells and in indirect contact. We also examined changes in biomaterials after interaction with cells, as well as the behavior of biomaterials in various conditions in vitro.

Ова реакција подразумева низ повезаних догађаја од којих коначно зависи да ли ће имплантирани материјал бити добро прихваћен и обавити своју примарну улогу. Сваки материјал за организам представља страно тело и иницира пролазну инфламаторну реакцију, чија дужина трајања и интензитет умногоме одређују његову даљу судбину. На то утиче хемијски састав материјала, величина и облик његових гранула, порозност, компактност, као и степен биодеградабилности. Тако, судбина материјала може кренути једним од два пута: ка доброј интеграцији са околним ткивом, подршци ћелијама са којима интерагује и, код биодеградабилних материјала, замени здравим, функционалним ткивом, или ће пак изазвати снажан и продужен инфламаторни одговор који резултује потпуном изолацијом материјала од остатка ткива дебелом фиброзном капсулом. In vivo испитивања одговора ткива на имплантирани биоматеријал подразумевају истраживања на различитим анималним моделима и у различитим ткивима, са ортопичним или ектопичним имплантацијама. У овом раду ће бити дат преглед наших in vivo истраживања на различитим моделима имплантација. Користили смо ортопичне моделе за формирање кости код пацова и кунића и моделе супкутаних имплантација различитих биоматеријала мишевима и пацовима. За анализу импланата и околног ткива у временским периодима раног и касног одговора коришћене су бројне методе као што су хистолошка бојења, хистоморфометрија, имунохистохемија, СЕМ, радиографске методе, анализа специфичне експресије гена и друге. In vivo анимални модели у сврху претклиничких испитивања важни су за добијање смерница за клиничку примену.
AB  - Today's approach to tissue repair and regeneration often involves application of different biomaterials, alone or in combination with cells and/or growth factors. All materials, regardless of whether they are of natural origin or synthesized, cause a reaction in the surrounding tissue after implantation. This reaction involves a series of related events on which ultimately depends whether the implanted material will be well accepted and perform its primary role. Each material represents a foreign body for the organism and initiates a transient inflammatory reaction, the duration and intensity of which largely determine materil's further fate. Inflammatory reaction is influenced by material's chemical composition, the size and shape of its granules, porosity, compactness, as well as the degree of material's biodegradability. Thus, the fate of the material can go one of two ways: towards good integration with the surrounding tissue, supporting the cells with which it interacts and, in the case of biodegradable materials, replacement by healthy, functional tissue, or it will cause a strong inflammatory response resulting in the complete isolation of the material from the rest of the tissue with thick fibrous capsule. In vivo research of tissue response to implanted biomaterial involves investigation in different animal models and in different tissues, with orthotopic or ectopic implantations. We used orthotopic models for bone formation in rats and rabbits and models of subcutaneous implantation of various biomaterials in mice and rats. Numerous methods were used for the analysis of implants and surrounding tissue in the time periods of early and late response, such as histological staining, histomorphometry, immunohistochemistry, SEM, radiographic methods, analysis of specific gene expression and others. In vivo animal models for the purpose of preclinical studies are important in order to obtain guidelines for clinical application.

Germanium-doped hydroxyapatite: Synthesis and characterization of a new substituted apatite

AB  - Hydroxyapatite (HAp) is the major component of all boney tissues in mammals. Because of this omnipresence in the living world, HAp possesses an exceptional biocompatibility. The downside of this omnipresence, however, comes in the form of its mild to moderate biological activities. One means of augmenting these activities involves the doping of HAp with foreign ions. Here, the first synthesis and characterization of HAp doped with germanium ions is being reported. Germanium was deliberately integrated into the crystal lattice of HAp in the form of germanate anions. Approximately two thirds of the germanate ions introduced into the hydrothermal solution got incorporated into the HAp lattice, yielding the approximate stoichiometry of Ca10-x(PO4)5.62+y(GeO3)0.38(OH)2-z. Germanates replaced the phosphates of stoichiometric HAp and induced the expansion of the HAp lattice both along the screw axis of the calcium ion hexagons and in the direction parallel to the basal plane. Simultaneously, the larger size and the triple valency of the germanate ion as compared to the smaller and trivalent phosphates prompted the bond distortion and charge compensation through defect formation, which reduced the crystallinity and increased the microstrain of the HAp lattice. Vibrational spectroscopic analyses corroborated these crystallographic effects by demonstrating the enhanced heterogeneity of the environments surrounding the active modes after germanate ions were incorporated into HAp. Conforming to La Châtelier's principle, this reduction of the crystallographic order increased the capacity of the material for integration of adventitious carbonates. However, the inclusion of germanate ions induced a partial shift of these carbonates to the hydroxyl channel sites, thus decreasing the ratio of the B-type carbonation to the A-type carbonation. Introduced into HAp, germanium acted as a superb regulator of the particle size and morphology, enhancing their fineness and uniformity. Inclusion of germanate ions also increased the electrophoretic mobility and hydrodynamic surface charge density of the particles by reducing their size and by inducing a more stochastic distribution of terminal ionic groups due to the bending of the crystal facets. Overall, the doping of HAp with germanate ions facilitated the production of narrowly dispersed nanorods with a moderately enhanced structural disorder and with a pronounced potential for the biomedical niche. © 2022 The Author(s)

Incorporation of a lactone moiety in the steroid nucleus, as in previously obtained 3β-acetoxy-17-oxa-17a-homoandrost-5-en-16-one (A) and 3β-hidroxy-17-oxa-17a-homoandrost-5-en-16-one (B), often results in enhanced anticancer properties. In this work, chitosan-based (Ch) nanoparticles were created and loaded with potent anticancer steroidal compounds, A and B. Changes to hormone receptor binding and cytotoxicity were then measured. In agreement with our previous results for A and B, A- and B-loaded Ch displayed cytotoxic properties against cancer cell lines. Both A-Ch and B-Ch showed activity toward estrogen negative breast cancer (MDA-MB-231) and androgen negative prostate cancer cell lines (PC-3). Greater selectivity toward cancer cells versus healthy lung fibroblast (MRC-5) was observed for B-Ch particles. Cell viability and cytotoxicity measurements after a recovery period indicate more robust recovery of healthy cells versus malignant cells. Compounds A and B or their Ch-encapsulated forms were shown to have negligible affinity for the ligand binding domain of estrogen receptor β or the androgen receptor in a fluorescent yeast screen, suggesting a lack of estrogenicity and androgenicity. Steroid-loaded chitosan nanoparticles display strong cytotoxicity towards MDA-MB-231 and PC-3 with a lack of hormone activity, indicating their safety and efficacy.

AB  - Over the past decades, development of engineering sciences has vastly contributed to advancements in medicine by production of numerous devices for diagnostics and treatment. In the middle of the 20th century, a new scientific field, biomedical engineering (BE), was established, which has developed into an extremely complex scientific discipline requiring a distinctive educational profile. Various study programs in BE have been established at universities around the world but also at several universities in Serbia. Also, intensive research in this field is performed at several scientific institutions in Serbia. In the present paper, short summaries of the research results of several groups of engineers and medical doctors are presented as an illustration of the wide field of BE research and possibilities of its application in diagnosis and therapy of various diseases.

AB  - Amorphous calcium phosphate (ACP) is a material of high interest for dentistry, orthopedics, and other biomedical sectors. Being intrinsically metastable, the process of transformation of ACP into a crystalline phase upon heating is of high relevance for the development of innovative bioceramics. Here we have first studied the thermal behavior of a citrate-stabilized ACP (Cit-ACP) also doped with fluoride ions (Cit-FACP) prepared at three different nominal Cit/Ca ratios (i.e. 4, 2, 1) by differential thermal analysis. Next, the physico-chemical features of the crystalline products as well as the in vitro cell response to the materials were investigated. A citrate and fluoride free ACP sample was also tested as the blank. We have found that the activation energy of crystallization of Cit-(F)ACP samples is lower in comparison to the blank ACP and this is influenced by the nominal Cit/Ca molar ratio. Interestingly, we have discovered that the thermal treatment of Cit-(F)ACP at 800 °C yields hydroxyapatite (HA) or fluorapatite (FHA) as the main products differently from blank ACP that, like most of the ACPs reported in the literature, yields β-tricalcium phosphate. This was attributed to the Ca/P ratio of Cit-(F)ACP, which is similar to HA. A study of the crystalline products has revealed that all the (F)HA samples were non-cytotoxic, and retained carbonate ions in the crystal structure despite the heat treatment that should have induced decarbonation. The morphology of the products is influenced by the nominal Cit/Ca ratio and the presence of fluoride, ranging from spherical nanoparticles to micrometric hexagonal rods. Overall, our results prove that the thermal crystallization of Cit-(F)ACP is markedly different from classic ACP based materials and the thermal treatment of Cit-(F)ACP represents an attractive route for producing pure bioactive HA ceramics.

Interestingly, we have discovered that the thermal treatment of Cit-(F)ACP at 800 °C yields hydroxyapatite (HA) or fluorapatite (FHA) as the main products differently from blank ACP that, like most of the ACPs reported in the literature, yields β-tricalcium phosphate. This was attributed to the Ca/P ratio of Cit-(F)ACP, which is similar to HA. A study of the crystalline products has revealed that all the (F)HA samples were non-cytotoxic, and retained carbonate ions in the crystal structure despite the heat treatment that should have induced decarbonation. The morphology of the products is influenced by the nominal Cit/Ca ratio and the presence of fluoride, ranging from spherical nanoparticles to micrometric hexagonal rods. Overall, our results prove that the thermal crystallization of Cit-(F)ACP is markedly different from classic ACP based materials and the thermal treatment of Cit-(F)ACP represents an attractive route for producing pure bioactive HA ceramics.

Cytotoxicity tests showed that there was no significant decrease in the survival of healthy MRC-5 cells exposed to composite samples without chitosan lactate, while there was an increase in the number of viable cells in the sample containing ChOL. These findings show that there was improved cell proliferation, differentiation and cell viability in the ChOL-containing sample, which makes ACP/TiO2/ChOL coating a good candidate for applications in medicine and stomatology.

AB  - The aim of this work was to investigate corrosion resistivity, bioactivity, and antibacterial activity of novel nano-amorphous calcium phosphate (ACP) potentially multifunctional composite coatings with and without chitosan oligosaccharide lactate (ChOL), ACP + ChOL/TiO2 and ACP/TiO2 ACP + ChOL/TiO2, respectively, on the titanium substrate. The coatings were obtained by new single-step in situ anodization of the substrate to generate TiO2 and the anaphoretic electrodeposition process of ACP and ChOL. The obtained coatings were around 300 ± 15 μm thick and consisted of two phases, namely, TiO2 and hybrid composite phases. Both ACP/TiO2 and ACP + ChOL/TiO2 have improved corrosion stability, whereas the ACP + ChOL/TiO2 coating showed better corrosion stability. It was shown that at the very start of the deposition process, the formation of the ChOL/TiO2 layer takes place predominantly, which is followed by the inclusion of ChOL into ACP with simultaneous growth of TiO2. This deposition mechanism resulted in the formation of strongly bonded uniform stable coating with high corrosion resistance. In vitro bioactivity was investigated by immersion of the samples in simulated body fluid (SBF). There is in-bone-like apatite formation on both ACP/TiO2 and ACP + ChOL/TiO2 surfaces upon immersion into SBF, which was proven by X-ray diffraction and Fourier transform infrared spectroscopy. While ACP/TiO2 shows no antibacterial activity, ACP + ChOL/TiO2 samples exhibited three- to fourfold decreases in the number of Staphylococcus aureus and Pseudomonas aeruginosa, respectively, after 420 min. The probable mechanism is binding ChOL with the bacterial cell wall, inhibiting its growth, altering the permeability of the cell membrane, and leading to bacterial death.

The analysis showed significant difference at the 5% level of confidence.

AB  - Kalcijum fosfati (CP), među kojima su amorfni kalcijum fosfat (ACP) i hidroksiapatit (HAp), zajedno sa slojevima Ti i TiO2, pronašli su ogromnu primenu u preventivnoj i regenerativnoj medicini zbog svoje odlične biokompatibilnosti, netoksičnih svojstava i sposobnosti da učestvuju u metaboličnim procesima organizama. U ovom radu je prikazana citotoksičnost multifunkcionalnih i hibridnih kompozitnih prevlaka amorfnog kalcijum fosfata (ACP) i hitozan laktata (ChOL) na ćelijskoj liniji humanih fibroblasta pluća MRC-5. ACP/TiO2 i ACP/TiO2/ChOL prevlake su taložene novim procesom in situ anodizacije/anaforetske depozicije pri konstantnom naponu od 60 V tokom 180 s na 25 ° C. Testovi citotoksičnosti pokazali su da nije bilo značajnog smanjenja preživljavanja zdravih ćelija MRC-5 u kompozitnim uzorcima Ti i ACP/TiO2, dok je došlo do povećanja broja viabilnih ćelija u uzorku ACP/TiO2/ChOL. Pokazano je poboljšanje ćelijske proliferacije, diferencijacije i viabilnosti ćelija. Uzorak koji sadrži ACP/TiO2/ChOL prevlaku pokazao je negativnu citotoksičnost u DET i u MTT testovima. Zabeležen je veći oporavak humanih ćelija fibroblasta pluća MRC-5 u poređenju sa kontrolnim uzorkom posle 48 h oporavka. Iz ovih rezultata može se zaključiti da multifunkcionalna kompozitna prevlaka ACP/TiO2/ChOL nije samo necitotoksična, već prisustvo ChOL-a u prevlaci poboljšava ćelijsku proliferaciju, diferencijaciju i viabilnost ćelija. Na osnovu dobijenih rezultata može se zaključiti da oba kompozitna materijala koji su korišćeni u studijama nisu citotoksični za korišćene ćelijske linije, te da 5 tež.% ChOL-a ima pozitivan efekat na netoksičnost materijala. Na osnovu prikazanih rezultata u ovom radu i prethodnih objavljenih rezultata fizičko-hemijskih i bioaktivnih svojstava kompozitnog materijala ACP/TiO2/ChOL, može se zaključiti da bi dalji razvoj, kao i potencijalne pretkliničke studije bili u velikoj meri opravdani.
AB  - Calcium phosphates (CP), amongst which amorphous calcium phosphate (ACP) and hydroxyapatite (HAp), along with Ti and TiO2 layers, have found vast applications in preventive and regenerative medicine due to their excellent biocompatibility, nontoxic properties and ability to participate in the normal metabolism of organisms. In this paper cytotoxicity of amorphous calcium phosphate (ACP) and chitosan lactate (ChOL) multifunctional and hybrid composite coatings on MRC-5 human lung fibroblast cell line is presented. ACP/TiO2 and ACP/TiO2/ChOL are deposited by coatings new in situ anodization/anaphoretic deposition process at constant voltage of 60 V for 180 s at 25 °C. Cytotoxicity tests showed that there was no significant decrease in the survival of healthy MRC-5 cells in the Ti and ACP/TiO2 composite samples, while there was an increase in the number of viable cells in the ACP/TiO2/ChOL sample. There is improved cell proliferation, differentiation and cell viability in the later. Sample containing ACP/TiO2/ChOL coating showed negative cytotoxicity in both DET and MTT tests. Greater recovery of MRC-5 human lung fibroblasts cells was observed compared to the control sample after 48 h of recovery. From these results, it can be concluded that not only the ACP/TiO2/ChOL multifunctional composite coating is non-cytotoxic, but the presence of ChOL in the coating improves cell proliferation, differentiation and cell viability. Based on the obtained results, it can be concluded that both composite materials used in the studies are non-cytotoxic to the cell lines used, and that 5 wt.% of ChOL has a positive effect on the non-toxicity of the material. Based on presented results in this paper and previous published results of the physicochemical and bioactive properties of the ACP/TiO2/ChOL composite material, it can be concluded that further development as well as potential preclinical studies would be largely justified.
