AB  - Objective: Acinetobacter baumannii is a pathogenic species which presents a danger to healthcare facilities due to the rapid spread of antibiotic resistance. Plants are currently being explored as a potential source of bioactive compounds that could be used to combat infectious diseases. Cinnamon is used as a food spice, but essential oil has been proven for antimicrobial activity. Due to reduced stability and solubility of essential oil, nanoemulsion (NE) synthesis could provide stronger antimicrobial effects. Investigation and comparison of antimicrobial activity of cinnamon (Cinnamomum zeylanicum L.) bark essential oil (EO) and NE on the A. baumannii ATCC19606 and clinical isolates. Effect of EO and NE on biofilm formation and biofilm eradication.
Methods: GC/MS was performed in order to determine chemical composition of commercially purchased EO (P0125285, Frey + Lau, GmbH, Henstedt-Ulzburg, Germany). Droplet/particle size and polydispersity index of NE was determined by photon correlation spectroscopy (PCS). Minimal inhibitory concentration of EO and NE were defined using MIC assay. Effects of EO and NE were also examined on biofilm formation and eradication. Crystal violet staining was used for biofilm biomass quantification.
Results: GC/MS analysis determined that the most common compound was trans-Cinnamaldehyde (61.9%). NE droplet/particles had multimodal distribution, shown by PCS. MIC values for EO were in range 0.25mg/mL — 0.5mg/mL, and for NE 0.125mg/mL — 0.25mg/mL. Both tested substances showed good effect on biofilm eradication, and destroyed biofilm biomass up to 64%, whilst the inhibition of biofilm formation was up to 70%.
Conclusion: Taking all the results into account, it is a good start for further investigations of both EO and NE as a potential antimicrobial agent. Deeper knowledge about the mechanisms of actions of both of these tested substances could help us to understand the best and the most effective way to use them, in order to combat against Acinetobacter baumannii.

AB  - Антимикробна резистенција представља велики проблем за јавно здравље због ограничених терапијских могућности. Acinetobacter baumannii је опортунистички патоген, отпоран на многе групе антибиотика и доводи до појаве болничких инфекција. Светска здравствена организација прогласила је A. baumannii за патогенa који ефикасно избегава ефекат конвенционалних антибиотика, тако да је потреба за новим антимикробним средствима евидентна. Природни производи укључујући етарска уља су препозната као добри антимикробни агенси. Цимет је вековима познат зачин који има широку примену и за етарско уље (ЕУ цимета) је показано да поседује различите биолошке активности. Циљ ове студије је било испитивање ефекта ЕУ цимета и његове новосинтетисане наноемулзије (НА) на формирање биофилма A. baumannii. ЕУ цимета је хемијски окарактерисано применом GC/MS анализе. Величина капи и полидисперзитет припремљених НА испитани су фотонском корелационом спектроскопијом. МИК вредности су одређене микродилуционом методом, а антибиофилм активност бојењем кристал виолетом. GC/MS анализа је показала да је најзаступљенија компонента испитиваног ЕУ транс-цинамалдехид (61,9%), док НА показују мултимодалну расподелу величине честица. МИК вредности су се кретале у опсегу 0,25-0,5 mg/mL за EУ и 0,125-0,25 mg/mL за НА. Инхибиција формирања биофилма се кретала до 59% и 69%, за ЕУ и НА. Добијени резултати показују да НА има јачи антимикробни и антибиофилм ефекат, па се може рећи да би емулзификација могла бити приступ ка ефикаснијем сузбијању инфекција А. baumannii.

AB  - Селен (Se) је металоид неопходан за правилно функционисање организма. Улази у састав више од 25 селенопротеина. Неопходан је за антиоксидативни систем (АОС), метаболизам тиреоидних хормона, синтезу ДНК, плодност, имунитет… Концентрације Se у организму варирају у малом опсегу од чега зависи његов дефицит, оптималан биолошки учинак или токсичност.1 Селен продире у организам у неорганском (селенит или селенат) и органском (селенометионин и селеноцистеин) облику. Органски облици селена показују најбољу биорасположивост.2 Биомедицински ефекти честица нано селена (SeNPs) су интензивно истраживани током протекле деценије.3 Циљ ове студије био је да се упореде ефекти SeNPs и Na-селенита на АОС у плаценти гравидних женки пацова. Плацента је посредник између мајке и фетуса и учествује у транспорту Se током целог интраутериног развоја. Гравидне женке Wistar albino пацова су гаважом свакога дана добијале SeNPs или Na-селенит. Испитивана је активност супероксид дисмутазе (SOD), каталазе (CAT), глутатион пероксидазе (GSH-Px), глутатион редуктазе (GR) и глутатион С-трансферазе (GST), као и концентрације глутатиона (GSH) и сулфхидрилних група (SH). SeNPs је довео до значајног повећања активности SOD, CAT и GSH-Px и смањења активности GST и концентрација GSH и SH у плаценти, док је третман Na-селенитом довео до повећања концентрације GSH у односу на контроле. Добијени резултати указују да SeNPs и Na-селенит изазивају различит физиолошки ефекат у плаценти гравидних женки пацова, а даља истраживања треба да утврде да ли је у питању повећано оксидативно оптерећење или побољшана антиоксидативна заштита.

From classical to machine learning aided approach - hydrothermal synthesis planning for metal oxide nanomaterials

AB  - In the past decade, scientists have made an outstanding progress in machine learning techniques, based on natural language processing, that takes advantage of vast number of material science documents to gain novel aspects on materials synthesis. Herein, we are comparing hydrothermal synthesis recipes for obtaining metal oxide nanomaterials carried out in our laboratory with learned synthesis recipes in terms of precursors, processing parameters, steps and product characteristics. As a starting point, open source tools and pre-trained models are used to get optimal hydrothermal synthesis recepies for desired material. This new approach is showing a great potential to become a standard method in planning viable synthesis routes for new materials.

As a starting point, open source tools and pre-trained models are used to get optimal hydrothermal synthesis recepies for desired material. AB  - Introduction: As a result of human activity, the amount of plastic waste has increased immensely in the environment. While the main concern has been orientated toward visible litter, more recently fragments of plastic less than 5 mm in diameter, called microplastic particles (MP), raised the attention due to their ability to induce toxic effects in organisms. The toxicity of MP increases with a decrease in size since smaller sized particles are absorbed systematically and accumulated in different organs/tissues, including brain.

Aim: Present study is designed to compare possible toxic acute effects of different sized MP, originating from polyethylene-terephthalate bottles, on toxicity signs, sensory-motor functions and relative brain weight in young male Wistar rats.

Methods: Rats (n = 24) were divided in four experimental groups: (I) intact (controls); Q that orally received a 2.5 ml of Milli-Q; P1/P2 that were respectively orally administrated with 35 mg/kg of MP in median diameter of either 40 or 70 μm dissolved in a 2.5 ml of Milli-Q. 24 h after treatments, using toxicity clinical test, we assessed the agitation, convulsion, piloerection, sleepiness and lethargy. Parameters were valued as: – (no effect), + (mild effect), ++ (moderate effect) and +++ (major effect). Then, we evaluated sensory-motor functions by monitoring: consciousness (scored 0/1 point), respiration (scored 0/1), spontaneous activity (scored 0/1/2/3), forepaw outstretching (scored 0/1/2), climbing (scored 0/1), visual placing (scored 0/1), cage grasp (scored 0/1), gait posture (scored 0/1/2), geotaxis (scored 0/1), hearing (scored 0/1) and pacing/circling (scored 0/1). Total score was expressed as the sum of average scores of all investigated parameters and graded on the scale from 0 to 15. Then, rats were weighed and sacrificed, while their brains were carefully removed and weighed. The relative brain weights were calculated as the ratio of brain (wet tissue, mg) weight to body weight (g). For toxicity clinical testing a descriptive analysis was used while other data were analysed by One-way analysis of variance followed by Tukey’s posthoc test. Results were considered significant at p < 0.05.
Results: Clinical toxicity signs were similar between groups while results of sensory-motor test point to unaltered sensory-motor functions upon MP treatments compared to controls (F (3, 20) = 0.9538; P = 0.4336). The absence of changes might be attributed to acute treatment, brief evaluation period and lack of cumulative effect upon MP treatments. Nonetheless, only smaller sized MP led to an increase in relative brain weight compared to controls (F (3, 20) = 3.425; P = 0.0369; p < 0.05) which might indicate the capability of smaller sized MP to cross blood-brain barrier and be accumulated in the brain inducing oxidative stress and inflammatory response. Although larger sized MP did not cause changes in relative brain weight, the possible modulations of additional parameters that were not the focus of this study should not be omitted.
Conclusion: According to the presented results, even a smaller sized MP applied in a single dose could be harmful to health, thus, greater control of plastic usage and disposal is essential for humanity.

AB  - Биофилм као животна форма представља један од најважнијих фактора вируленције који је заступљен у 80-90% инфекција, а посебно на материјалима који се користе у ортопедији. Бактерије у биофилму су вишеструко отпорније на антибиотике и спољашње факторе, те је превенција развоја биофилма некада једини ефикасан метод спречавања појаве и ширења инфекције. У последње време интензивно се проучавају наноструктуирани материјали отпорни на колонизацију бактерија и развој биофилма. Селен је један од елемената који у својој наноформи показује добре биолошке ефекте, а ресвератрол је популарни полифенол чији се потенцијал за примену у медицини интензивно истражује. Добијањем обе ове компоненте у нано- облицима, отварају се даље могућности комбиновања и стварања композитног материјала са побољшаним особинама, за примену у превенцији биофилма. Ресвератрол и наночестице селена појединачно показују добар, али недовољно истражен антимикробни потенцијал. Након њиховог спајања испитали смо ефекат добијеног композита у спречавању формирања биофилма код више бактеријских врста. Концентрације композита од 6% (и ниже) су доводиле до инхибиције формирања биофилма од најмање 20% код већине тестираних бактерија. Посебно се истиче добар антибиофилм ефекат на сојевима P. aeruginosa, где је на тестираним концентрацијама уочена инхибиција формирања биофилма у опсегу од 20-60%. Такође, антибиофилм ефекат композита је у већини случајева бољи у односу на саме честице у истим концентрацијама.

AB  - Selenium is an especially important element for the functioning of antioxidative enzymes, making it one of the essential micronutrients. Although selenium is required in extremely small amounts, often it is lacking from the organism. However, inorganic and organic sources of selenium are known to possess considerable toxicity. Recently, the nanoparticle form of selenium has been shown to exhibit lesser toxicity, and also better antimicrobial and other biological effects, than selenium-containing compounds. Nanotechnology has already found application in many fields, for improving material properties for various purposes, among them, in the process of improving food characteristics. It is possible that selenium nanoparticles can be applied as antimicrobial agents in food, and there has been growing interest in this area of research. We have synthesized selenium nanoparticles (SeNPs) by chemical reduction method, using sodium selenite, ascorbic acid, and bovine serum albumin. The antimicrobial activity of these particles has been assessed by minimum inhibitory concentration assay and colony-forming units count, on Listeria monocytogenes and Salmonella enterica, which are common bacterial food contaminants. As intestinal microbiota, especially probiotic bacteria, have been shown to play important role in organism functioning, we also tested how SeNPs influence Lactobacillus rhamosus and Lactobacillus plantarum. To evaluate cytotoxicity, we used MTT assay on the human intestinal cell line (HT-29). Antioxidative properties were assessed by DPPH assay. SeNPs showed higher antibacterial activity on pathogenic bacteria than on probiotic strains. Cytotoxic effect was present only in the highest tested concentrations. The antioxidative activity was considerable, resulting in above 80% reduction of DPPH free radical, at 1 g/ml of SeNPs. Based on these results, SeNPs are a promising subject for further research in developing materials for food packaging or as antimicrobial selenium additives.

AB  - One of the major issues related to medical devices and especially urinary stents are infections caused by different strains of bacteria and fungi, mainly in light of the recent rise in microbial resistance to existing antibiotics. Lately, it has been shown that nanomaterials could be superior alternatives to conventional antibiotics. Generally, nanoparticles are used for many applications in the biomedical field primarily due to the ability to adjust and control their physicochemical properties as well as their great reactivity due to the large surface-to-volume ratio. This has led to the formation of a new research field called nanomedicine which can be defined as the use of nanotechnology and nanomaterials in diagnostics, imaging, observing, prevention, control, and treatment of diseases. For example, coverings or coatings based on nanomaterials are now seen as a promising strategy for preventing or treating biofilms formation on healthcare kits, implants, and medical devices. Toxicity, inappropriate delivery, or degradation of conventionally used drugs for the treatment of infections may be avoided by using nanoparticles without or with encapsulated/immobilized active substances. Most of the materials which are used and examined for the preparation of the nanoparticles with encapsulated/immobilized active substances or smart reactive nanomaterials with antimicrobial effects are polymers, naturally derived antimicrobials, metal-based and non-metallic materials. This chapter provides an overview of the current state and future perspectives of the nanoparticle-based systems based on these materials for prevention, control, or elimination of biofilm-related infections on urinary stents. It also addresses manufacturing conditions indicating the huge potential for the improvement of existing and development of new promising stent solutions.

This has led to the formation of a new research field called nanomedicine which can be defined as the use of nanotechnology and nanomaterials in diagnostics, imaging, observing, prevention, control, and treatment of diseases. For example, coverings or coatings based on nanomaterials are now seen as a promising strategy for preventing or treating biofilms formation on healthcare kits, implants, and medical devices. Toxicity, inappropriate delivery, or degradation of conventionally used drugs for the treatment of infections may be avoided by using nanoparticles without or with encapsulated/immobilized active substances. Most of the materials which are used and examined for the preparation of the nanoparticles with encapsulated/immobilized active substances or smart reactive nanomaterials with antimicrobial effects are polymers, naturally derived antimicrobials, metal-based and non-metallic materials. AB  - Acinetobacter baumannii is an emerging nosocomial pathogen resistant to a wide spectrum of antibiotics, with great potential to form a biofilm, which further aggravates treatment of infections caused by it. Therefore, searching for new potent agents that are efficient against A. baumannii seems to be a necessity. One of them, which has already been proven to possess a wide spectrum of biological activities, including antimicrobial effect, is cinnamon essential oil. Still, further increase of antibacterial efficacy and improvement of bioavailability of cinnamon oil is possible by emulsification process. The aim of this study was comparative analysis of cinnamon essential oil and its emulsion against biofilm forming A. baumannii clinical isolates. Furthermore, the investigation of toxicological aspects of possible applications of essential oil and emulsion was done as well. Gas chromatography–mass spectrometry of essential oil indicated trans-cinnamaldehyde as the most abundant component. The cinnamon emulsion was synthesized from cinnamon essential oil by combining modified low- and high- energy methods. Synthesized emulsion was characterized with Fourier-transform infrared spectroscopy and photon correlation spectroscopy. Both substances exhibited significant antibacterial (minimal inhibitory concentrations in the range 0.125–0.5 mg/ml) and antibiofilm effects (inhibitions of formation and reduction of pre-formed biofilm were 47–81 and 30–62%, respectively). Compared to essential oil, the efficacy of emulsion was even stronger considering the small share of pure oil (20%) in the emulsion. The result of biofilm eradication assay was confirmed by scanning electron microscopy. Even though the cytotoxicity was high especially for the emulsion, genotoxicity was not determined. In conclusion, strong antibacterial/antibiofilm effect against A. baumannii of the cinnamon essential oil and the fact that emulsification even potentiated the activity, seems to be of great significance. Observed cytotoxicity implicated that further analysis is needed in order to clearly determine active principles being responsible for obtained antibacterial/antibiofilm and cytotoxic properties. AB  - Multidrug-resistant bacterial strains represent an emerging global health threat and a great obstacle for bone tissue engineering. One of the major components of the extracellular matrix of the bone is a collagen protein, while selenium is an element that has antimicrobial potential, and is also important for bone metabolism and bone health. Here we represent the incorporation of selenium nanoparticles (SeNPs) synthesized by the green chemical reduction method into collagen gels to produce a composite material, collagen/SeNPs, with antimicrobial properties. The samples were comprehensively characterized by zeta potential measurements, dynamic light scattering inductively coupled plasma-mass spectrometry (ICP-MS), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), optical microscopy, field-emission scanning electron microscopy (FE-SEM), and differential scanning calorimetry The cytotoxicity of the SeNPS, as well as collagen/SeNPs, was tested on the MRC-5 cells. It was revealed that collagen/SeNPS expressed a lower cytotoxic effect. Collagen/SeNPs showed significant antibacterial activity against all tested Gram-positive strains, the major causative agents of orthopedic infections as well as Candida albicans. Furthermore, three-dimensional β-tricalcium phosphate (3D-TCP) scaffolds were fabricated by a well-established 3D printing (lithography) method, and afterward preliminary coated by newly-synthesized SeNPs or collagen/SeNPs. In addition, uncoated 3D-TCP scaffolds as well as coated by collagen/SeNPs were subjected to biofilm formation. The production of Staphylococcus aureus biofilm on coated scaffolds by collagen/SeNPs was significantly reduced compared to the uncoated ones. © The Author(s) 2022.

AB  - The chemical element selenium (Se) is a nonmetal that is in trace amounts indispensable for normal cellular functioning. During pregnancy, a low Se status can increase the risk of oxidative stress. However, elevated concentrations of Se in the body can also cause oxidative stress. This study aimed to compare the effects of BSA-stabilized Se nanoparticles (SeNPs, Se0) (BSA-bovine serum albumin) and inorganic sodium selenite (NaSe, Se+4) supplementation on the histological structure of the placenta, oxidative stress parameters and the total placental Se concentration of Wistar rats during pregnancy. Pregnant females were randomized into four groups: (i) intact controls; (ii) controls that were dosed by daily oral gavage with 8.6% bovine serum albumin (BSA) and 0.125 M vit C; (iii) the SeNP group that was administered 0.5 mg of SeNPs stabilized with 8.6% BSA and 0.125 M vit C/kg bw/day by oral gavage dosing; (iv) the NaSe group, gavage dosed with 0.5 mg Na2SeO3/kg bw/day. The treatment of pregnant females started on gestational day one, lasted until day 20, and on day 21 of gestation, the fetuses with the placenta were removed from the uterus. Our findings show that the mode of action of equivalent concentrations of Se in SeNPs and NaSe depended on its redox state and chemical structure. Administration of SeNPs (Se0) increased fetal lethality and induced changes in the antioxidative defense parameters in the placenta. The accumulation of Se in the placenta was highest in SeNP-treated animals. All obtained data indicate an increased bioavailability of Se in its organic nano form and Se0 redox state in comparison to its inorganic sodium selenite form and Se+4 redox state.

AB  - The chemical element selenium (Se) is a nonmetal that is in trace amounts indispensable for normal cellular functioning. During pregnancy, a low Se status can increase the risk of oxidative stress. However, elevated concentrations of Se in the body can also cause oxidative stress. This study aimed to compare the effects of BSA-stabilized Se nanoparticles (SeNPs, Se0) (BSA-bovine serum albumin) and inorganic sodium selenite (NaSe, Se+4) supplementation on the histological structure of the placenta, oxidative stress parameters and the total placental Se concentration of Wistar rats during pregnancy. Pregnant females were randomized into four groups: (i) intact controls; (ii) controls that were dosed by daily oral gavage with 8.6% bovine serum albumin (BSA) and 0.125 M vit C; (iii) the SeNP group that was administered 0.5 mg of SeNPs stabilized with 8.6% BSA and 0.125 M vit C/kg bw/day by oral gavage dosing; (iv) the NaSe group, gavage dosed with 0.5 mg Na2SeO3/kg bw/day. The treatment of pregnant females started on gestational day one, lasted until day 20, and on day 21 of gestation, the fetuses with the placenta were removed from the uterus. Our findings show that the mode of action of equivalent concentrations of Se in SeNPs and NaSe depended on its redox state and chemical structure. Administration of SeNPs (Se0) increased fetal lethality and induced changes in the antioxidative defense parameters in the placenta. The accumulation of Se in the placenta was highest in SeNP-treated animals. All obtained data indicate an increased bioavailability of Se in its organic nano form and Se0 redox state in comparison to its inorganic sodium selenite form and Se+4 redox state.

AB  - Antimicrobial resistance (AMR) and infections caused by multidrug-resistant microbial pathogens represent one of the major clinical challenges responsible for high-level morbidity and mortality. They are a significant problem to the public health and the economic stability of societies all over the world. In November 2021 WHO has declared AMR as one of the top 10 global public health threats (Antimicrobial resistance (who.int)). According to CDC’s 2019 AMR Report, although declining since 2013, with 2.8 million new cases and more than 35,000 deaths each year, the number of people facing the AMR problem in United States is still too high (Antibiotic Resistance Threats in the United States, 2019 (cdc.gov)). AMR also remains the major health concern of EU with more than 670,000 new cases of infections caused by antibiotic-resistant bacterial strains and more than 33,000 deaths per year (Antimicrobial resistance surveillance in Europe 2022 - 2020 data (europa.eu)). In 2019 China recorded 39-% drop of antibiotic use in hospitalised patients compare to 2011 (Antimicrobial resistance - China (who.int)). Still, with 73,000 estimated new cases only for multi-drug resistant tuberculosis, the region remained at second position of the global highest incident rates (Antimicrobial resistance - China (who.int)). Each year a large number of people receive different kinds of implants, for example, hip or knee. However recent discoveries reveal that, either during the operative protocol or due to secondary infections, the implant’s surface could be colonized by bacteria, fungi, or both which can have serious consequences on a patient’s health. According to Annual Epidemiological Report of ECDC in 2016, post-surgical infections were identified as most common healthcare-associated infections (Surgical site infections - Annual Epidemiological Report 2016 [2014 data] (europa.eu)). In recent years it has been also recognized that microbial biofilms are ubiquitous, which has resulted in a number of studies from a biofilm perspective. Currently, great efforts are focused on the development of innovative therapeutic strategies regarding both novel drug candidates and drug delivery systems for treating microbial infections associated with implants. However, despite all these efforts as well as the urgent need, an effective and long-lasting solution to this problem is still not found. In the last decades, great attention is paid to nanostructured polymeric materials and nanocomposites because of their unique properties, which make them appropriate candidates for various applications in different medical and pharmaceutical fields. This Research Topic draws attention to the up-to-date findings regarding these issues and advanced therapeutic strategies and approaches as possible solutions.

AB  - Natural chemicals and earth elements are increasingly used in research as basis for novel materials intended for use in medicine. Among phytopharmaceuticals, more specifically polyphenols, resveratrol is known for its’ antioxidative, anticancer, antimicrobial, and other beneficial effects. Selenium, an essential trace element, is lately being recognized in nanoparticle form as less toxic and equally or more efficient than commercially available forms. The synergy of these two agents have not been shown until lately, when their synergistic antioxidative and gene expression effects were investigated for the purpose of treating Alcheimer disease. During our previous research, we have successfully synthesized pure resveratrol particles, as well as selenium nanoparticles (SeNPs). Both of them were separately investigated regarding their biological activities. The first step in evaluation of their possible synergistic antioxidative effect was obtaining the stable composite of these two materials. Synthesis parameters and processing methods were varied, and obtained suspensions assessed by their macroscopic and microscopic characteristics. The sample with both components homogeneously distributed in the particle form, was chosen for further experiments. Ultraviolet-visible (UV-Vis) spectrophotometry and Fourier transform infrared spectroscopy (FTIR) were used to characterize sample, and antioxidative activity (by DPPH reduction assay), and cytocompatibility (using MTT cytotoxicity assay) were additionally determined. Results showed improved cytocompatibility as compared to pure resveratrol particles, and preserved, significantly high antioxidative potential.

Also, whole-genome sequencing (WGS) and gene expression analyses were used to elucidate the mechanisms of colistin resistance. Exceptionally strong synergistic activity (FICI range 0.004–0.035) of colistin and SeNPs against colistin- resistant isolates was revealed. Colistin (0.5 or 1 μg ml −1 ) used in combination with SeNPs (0.5 μg ml−1 ) was able to reduce initial inoculum during the first 4 h of incuba- tion, in contrast to colistin (0.5, 1 or 2 μg ml−1 ) alone. Conclusions: These findings propose colistin/SeNPs combination as a new option to fight PDR Ac. baumannii, the therapeutic possibilities of which should be proved in future in vivo studies. Significance and Impact of Study: Here we present the first evidence of synergy between colistin and selenium compounds against bacteria in general. Also, WGS and gene expression analyses provide some new insights into Ac. baumannii colistin resistance mechanisms.

Methods and Results: Chequerboard and time-kill assays were employed to ex- plore the potential synergistic interactions between colistin and SeNPs against Ac. baumannii isolates (8), previously determined as colistin-resistant (MIC range 16– 256 μg ml−1 ). Also, whole-genome sequencing (WGS) and gene expression analyses were used to elucidate the mechanisms of colistin resistance. Exceptionally strong synergistic activity (FICI range 0.004–0.035) of colistin and SeNPs against colistin- resistant isolates was revealed. Colistin (0.5 or 1 μg ml −1 ) used in combination with SeNPs (0.5 μg ml−1 ) was able to reduce initial inoculum during the first 4 h of incuba- tion, in contrast to colistin (0.5, 1 or 2 μg ml−1 ) alone. Conclusions: These findings propose colistin/SeNPs combination as a new option to fight PDR Ac. baumannii, the therapeutic possibilities of which should be proved in future in vivo studies. Significance and Impact of Study: Here we present the first evidence of synergy between colistin and selenium compounds against bacteria in general. Also, WGS and gene expression analyses provide some new insights into Ac. baumannii colistin resistance mechanisms.

AB  - Resveratrol, polyphenol from stilbene family, has demonstrated beneficial effects on human health in numerous studies. Among these effects are cardioprotective, anti-cancer, neuroprotective, antimicrobial and other. In vivo, application of this phytochemical often lacks significant results. This is mainly because of its poor bioavailability, caused by low water solubility, sensitivity to light, oxygen and pH change, and high reactivity. Micro- and nanoformulations of resveratrol have been described to highly improve bioavailabiliy. However, there are limitations to this approach, such as demanding synthesis process, necessity of expensive or toxic chemicals, or poor loading capacity. We have used a simple physicochemical solvent-non solvent method to synthetise additive-free, stabile and uniform elongated resveratrol micro- and nanoparticles (RES-particles). The drying conditions were varied to examine their influence on the morphological characteristics of resveratrol particles. Differential effects of drying at ambient, low, and elevated temperature as well as effects of centrifugation and stability of RES-particle suspension during the time were documented. RES-particles were characterized by X-ray diffraction (XRD), differential scanning calorimetry (DSC), ultraviolet-visible (UV-Vis) spectrophotometry, Fourier transform infrared spectroscopy (FTIR) and optical microscopy. Biocompatibility of RESparticles was preliminary assessed through MTT toxicity assay.

AB  - An increasing lack of available therapeutic options against Acinetobacter baumannii urged researchers to seek alternative ways to fight this extremely resistant nosocomial pathogen. Targeting its virulence appears to be a promising strategy, as it offers considerably reduced selection of resistant mutants. In this study, we tested antibiofilm potential of four synthetic chalcone derivatives against A. baumannii. Compound that showed the greatest activity was selected for further evaluation of its antivirulence properties. Real-time PCR was used to evaluate mRNA expression of biofilm-associated virulence factor genes (ompA, bap, abaI) in treated A. baumannii strains. Also, we examined virulence properties related to the expression of these genes, such as fibronectin- and collagen-mediated adhesion, surface motility, and quorum-sensing activity. The results revealed that the expression of all tested genes is downregulated together with the reduction of adhesion and motility. The conclusion is that 2′-hydroxy-2-methoxychalcone exhibits antivirulence activity against A. baumannii by inhibiting the expression of ompA and bap genes, which is reflected in reduced biofilm formation, adhesion, and surface motility.

AB  - We report an innovative low-cost wet  precipitation  synthesis  method  for  gelatin-modified   zinc oxide nanoparticles (GM ZnO NPs) at the inter- face  between  the  gelatin  hydrogel  and  aqueous  elec- trolyte.  Diffusion  of  ammonia  through  the  hydrogel   matrices with different gelatin contents induced pre- cipitation  of  the  product  in  contact  with  the  surface   of  the  aqueous  solution  of  zinc  ions.  The  obtained   precipitate  was  subjected  to  thermal  treatment  to  partially  decompose  the  adsorbed  gelatin  in  the  NP   structure. Physicochemical properties of obtained  GM  ZnO  NPs  were  characterized  by  X-ray  powder   diffraction (XRD), scanning electron microscopy  (SEM), Fourier transform infrared spectroscopy  (FTIR), differential thermal analysis (DTA), thermo- gravimetry (TG), photon correlation spectroscopy  (PCS),  zeta  potential  measurements,  and  inductively   coupled  plasma-mass  spectrometry  (ICP-MS).  The   estimated mean crystallite size of GM ZnO NP pow- ders was in the range from 5.8 to 12.1 nm. The syn- thesized  NPs  exhibited  nanosheet  morphology  and   arranged into flake-like aggregates. The toxic poten- tial was investigated in vitro in human hepatocellular  carcinoma cell line HepG2. The thiazolyl blue tetra- zolium bromide (MTS) assay was used to assess cell  viability,  2′,7′-dichlor-fluorescein-diacetate  (DCFH- DA)  assay  to  examine  the  formation  of  intracellu- lar  reactive  oxygen  species  (ROS),  and  comet  assay   to  evaluate  the  genotoxic  response.  GM  ZnO  NPs   slightly reduced HepG2 cell viability, did not induce  ROS formation, and showed low genotoxic potential  at  very  high  doses  (100  μg    mL−1).  ZnO  NPs  fabri- cated  and  modified  using  the  proposed  methodol- ogy deserve further study as potential candidates for  antibacterial agents or dietary supplements with low  overall toxicity.

AB  - Although selenium nanoparticles (SeNPs) have gained attention in the scientific community mostly through investigation of their anticancer activity, a great potential of this nanomaterial was recognized recently regarding its antimicrobial activity. The particle form, size, and surface chemistry have been recognized as crucial parameters determining the interaction of nanomaterials with biological entities. Furthermore, considering a narrow boundary between beneficial and toxic effects for selenium per se, it is clear that investigations of biomedical applications of SeNPs are very demanding and must be done with great precautions. The goal of this work is to evaluate the effects of SeNPs surface chemistry and structure on antimicrobial activity against several common bacterial strains, including Staphylococcus aureus (ATCC 6538), Enterococcus faecalis (ATCC 29212), Bacillus subtilis (ATCC 6633), and Kocuria rhizophila (ATCC 9341), as well as Escherichia coli (ATCC 8739), Salmonella Abony (NCTC 6017), Klebsiella pneumoniae (NCIMB 9111) and Pseudomonas aeruginosa (ATCC 9027), and the standard yeast strain Candida albicans (ATCC 10231). Three types of SeNPs were synthesized by chemical reduction approach using different stabilizers and reducing agents: (i) bovine serum albumin (BSA) + ascorbic acid, (ii) chitosan + ascorbic acid, and (iii) with glucose. A thorough physicochemical characterization of the obtained SeNPs was performed to determine the effects of varying synthesis parameters on their morphology, size, structure, and surface chemistry. All SeNPs were amorphous, with spherical morphology and size in the range 70–300 nm. However, the SeNPs obtained under different synthesis conditions, i.e. by using different stabilizers as well as reducing agents, exhibited different antimicrobial activity as well as cytotoxicity which are crucial for their applications. In this paper, the antimicrobial screening of the selected systems is presented, which was determined by the broth microdilution method, and inhibitory influence on the production of monomicrobial and dual-species biofilm was evaluated. The potential mechanism of action of different systems is proposed. Additionally, the cytotoxicity of SeNPs was examined on the MRC-5 cell line, in the same concentration interval as for antimicrobial testing. It was shown that formulation SeNPs-BSA expressed a significantly lower cytotoxic effect than the other two formulations.

AB  - We report an innovative low-cost wet  precipitation  synthesis  method  for  gelatin-modified   zinc oxide nanoparticles (GM ZnO NPs) at the inter- face  between  the  gelatin  hydrogel  and  aqueous  elec- trolyte.  Diffusion  of  ammonia  through  the  hydrogel   matrices with different gelatin contents induced pre- cipitation  of  the  product  in  contact  with  the  surface   of  the  aqueous  solution  of  zinc  ions.  The  obtained   precipitate  was  subjected  to  thermal  treatment  to  partially  decompose  the  adsorbed  gelatin  in  the  NP   structure. Physicochemical properties of obtained  GM  ZnO  NPs  were  characterized  by  X-ray  powder   diffraction (XRD), scanning electron microscopy  (SEM), Fourier transform infrared spectroscopy  (FTIR), differential thermal analysis (DTA), thermo- gravimetry (TG), photon correlation spectroscopy  (PCS),  zeta  potential  measurements,  and  inductively   coupled  plasma-mass  spectrometry  (ICP-MS).  The   estimated mean crystallite size of GM ZnO NP pow- ders was in the range from 5.8 to 12.1 nm. The syn- thesized  NPs  exhibited  nanosheet  morphology  and   arranged into flake-like aggregates. The toxic poten- tial was investigated in vitro in human hepatocellular  carcinoma cell line HepG2. The thiazolyl blue tetra- zolium bromide (MTS) assay was used to assess cell  viability,  2′,7′-dichlor-fluorescein-diacetate  (DCFH- DA)  assay  to  examine  the  formation  of  intracellu- lar  reactive  oxygen  species  (ROS),  and  comet  assay   to  evaluate  the  genotoxic  response.  GM  ZnO  NPs   slightly reduced HepG2 cell viability, did not induce  ROS formation, and showed low genotoxic potential  at  very  high  doses  (100  μg    mL−1).  ZnO  NPs  fabri- cated  and  modified  using  the  proposed  methodol- ogy deserve further study as potential candidates for  antibacterial agents or dietary supplements with low  overall toxicity.

The considerable concern regarding bacterial resistance to antimicrobial agents, bacterial adhesion, and proliferation has generated intense research in this field. Although urgently needed, an effective and long-lasting solution to this problem, i.e. microbial and especially bacterial colonization, has not yet been found. In the last decades, nanomaterials have attracted much attention because of their unique physical, chemical, and mechanical properties, due to their high surface area and size at the nanoscale. Owing to these extraordinary characteristics, nanomaterials are appropriate candidates for various applications in different fields such as medicine, pharmacy, food industry, etc. New strategies based on the use of nanomaterials such as nanoparticles, nanocapsules, nanogels, nanofibers, nanocomposites, or nanocoatings for controlling microbial virulence factors are very promising. In addition special focus recently is paid on nanostructured polymeric materials and nanocomposites with antimicrobial properties. The increasing attention within the scientific community regarding this topic can be easily assessed by searching the suitable keywords in some recognized literature databases. According to Scopus, in the last 10 years, more than 1,300 peer-reviewed documents have been published jointly containing keywords such as “nanocomposites” and “antimicrobial”. Interestingly, in the more specified search which refers to the utilization of nanostructured polymeric materials, it could be observed a similar profile i.e. growing tendency.

AB  - An increasing lack of available therapeutic options against Acinetobacter baumannii urged researchers to seek alternative ways to fight this extremely resistant nosocomial pathogen. Targeting its virulence appears to be a promising strategy, as it offers considerably reduced selection of resistant mutants. In this study, we tested antibiofilm potential of four synthetic chalcone derivatives against A. baumannii. Compound that showed the greatest activity was selected for further evaluation of its antivirulence properties. Real-time PCR was used to evaluate mRNA expression of biofilm-associated virulence factor genes (ompA, bap, abaI) in treated A. baumannii strains. Also, we examined virulence properties related to the expression of these genes, such as fibronectin- and collagen-mediated adhesion, surface motility, and quorum-sensing activity. The results revealed that the expression of all tested genes is downregulated together with the reduction of adhesion and motility. The conclusion is that 2′-hydroxy-2-methoxychalcone exhibits antivirulence activity against A. baumannii by inhibiting the expression of ompA and bap genes, which is reflected in reduced biofilm formation, adhesion, and surface motility.

Protective Effect of an Exopolysaccharide Produced by Lactiplantibacillus plantarum BGAN8 Against Cadmium-Induced Toxicity in Caco-2 Cells

AB  - Cadmium (Cd) ranks seventh on the list of most significant potential threats to human health based on its suspected toxicity and the possibility of exposure to it. It has been reported that some bacterial exopolysaccharides (EPSs) have the ability to bind heavy metal ions. We therefore investigated the capacity of eight EPS-producing lactobacilli to adsorb Cd in the present study, and Lactiplantibacillus plantarum BGAN8 was chosen as the best candidate. In addition, we demonstrate that an EPS derived from BGAN8 (EPS-AN8) exhibits a high Cd-binding capacity and prevents Cd-mediated toxicity in intestinal epithelial Caco-2 cells. Simultaneous use of EPS-AN8 with Cd treatment prevents inflammation, disruption of tight-junction proteins, and oxidative stress. Our results indicate that the EPS in question has a strong potential to be used as a postbiotic in combatting the adverse effects of Cd. Moreover, we show that higher concentrations of EPS-AN8 can alleviate Cd-induced cell damage.

AB  - Nanočestice selena (SeNPs) su specifična forma ovog elementa koja je u poslednje vreme postala predmet brojnih istraživanja, pogotovo u oblasti biomedicine. Do sada je razvijeno više metoda sinteze SeNPs, među kojima se najčešće koriste one koje podrazumevaju redukciju iz njegovih jedinjenja. U ovom radu ispitan je uticaj dva stabilizatora na morfologiju, veličinu i kristalno uređenje SeNPs dobijenih redukcijom natrijum selenita askorbinskom kiselinom. Kao stabilizatori SeNPs korišćeni su goveđi serum albumin (BSA) ili poliglutaminska kiselina (PGA). Na osnovu rezultata dobijenih metodama skenirajuće elektronske mikroskopije (FESEM), transmisione elektronske mikroskopije (TEM), energetske disperzione spektroskopije (EDS), elektronske difrakcije sa selektovane površine (SAED) i merenjem zeta potencijala, utvrđeno je da mehanizam stabilizacije tj. izbor stabilizatora može prouzrokovati različito kristalno uređenje unutar SeNPs. BSA se pokazao kao efikasniji stabilizator jer omogućuje dobijanje uniformnijih, manjih i amorfnih nanočestica selena.
AB  - Selenium nanoparticles (SeNPs) are specific form of this element that has recently become the subject of numerous research, especially in the field of biomedicine. Several synthesis procedures for obtaining SeNPs have been developed so far, among those including reduction of selenium salts are the most frequently used. In this work, it is examined the effect of two stabilizing agents on morphology, size, and crystallinity of obtained SeNPs. For this purpose, bovine serum albumin (BSA) and polyglutamic acid (PGA) were used as stabilizing agents while reduction of sodium selenite with ascorbic acid was elected as a synthesis procedure. Based on the results obtained from scanning electron microscopy (FESEM), transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDS), selected area electron diffraction (SAED), and measurements of zeta potential, it was determined that the mechanism of stabilization i.e. choice of stabilizing agent can promote different crystalline arrangement within SeNPs. The BSA proved as a more effective stabilizing agent for SeNPs, as it provides obtaining the smaller, more uniform, and amorphous nanoparticles.
