AB  - Indications exist that paramagnetic calcium phosphates may be able to promote regeneration of bone faster than their regular, diamagnetic counterparts. In this study, analyzed was the influence of paramagnetic cobalt-substituted hydroxyapatite nanoparticles on osteoporotic alveolar bone regeneration in rats. Simultaneously, biocompatibility of the material was tested in vitro, on osteoblastic MC3T3-E1 and epithelial Caco-2 cells in culture. The material was shown to be biocompatible and nontoxic when added to epithelial monolayers in vitro, while it caused a substantial decrease in the cell viability as well as deformation of the cytoskeleton and cell morphology when incubated with the osteoblastic cells. In the course of 6 months after the implantation of the material containing different amounts of cobalt, ranging from 5 to 12 wt%, in the osteoporotic alveolar bone of the lower jaw, the following parameters were investigated: histopathological parameters, alkaline phosphatase and alveolar bone density. The best result in terms of osteoporotic bone tissue regeneration was observed for hydroxyapatite nanoparticles with the largest content of cobalt ions. The histological analysis showed a high level of reparatory ability of the nanoparticulate material implanted in the bone defect, paralleled by a corresponding increase in the alveolar bone density. The combined effect of growth factors from autologous plasma admixed to cobalt-substituted hydroxyapatite was furthermore shown to have a crucial effect on the augmented osteoporotic bone regeneration upon the implantation of the biomaterial investigated in this study.

AB  - In recent years, calcium hydroxyapatite (HAp) and its substituents are increasingly used in dentistry and medicine. The influence of nanoparticles Ca / Co-HAp in strengthening weak osteoporotic bone jaw tegmenta was tested in an experimental model. The study was conducted on Wistar soy rats, aged 6-8 weeks. The biomaterial was implanted in the osteoporoticly weakened mandible of these animals. The best results in the strengthening of the lower jaw bone tegmenta were achieved 24 weeks after implantation of hydroxyapatite nanoparticles in which the calcium ion was substituted with 12% of cobalt ions. Histochemical parameters of bone synthesis were in a statistically significant increase. SEM analysis showed a high degree of osteogenetic ability of nano-particulate material implanted in the bone defect.

AB  - Increased life expectancy in developed countries leads to an increase in the number of musculoskeletal disorders, such as osteoporosis, oasteoratritis thus compromising good dental treatment. There are many drug delivery systems based on hydroxyapatite used in bone tissue regeneration. Vitamin D3 is osteodiferentiation factor which regulates bone formation by increasing osteoblast differentiation and bone mineralization. The aim of this study is to examine new multifunctional nanoparticulate system for local delivery of active form of vitamin D3 by biochemical blood marker and histochemical analysis. The research was carried out on female Westar rats, aged 6-8 weeks, which have been implanted biomaterials in the artificial bone defect. Biochemical markers of osteogenesis were statistically significant after only 6 weeks of implantation. ALP activity in bone tissue was showed by histochemical analysis as well as high level reparatory skills. Local realized Vitamin D3 contribute to bone formation by increasing osteoblast differentiation and bone mineralization.

Good results in regeneration of reduced bone density were achieved in six weeks after implantation of the nanoparticles with paramagnetic properties. Biochemical markers of osteogenesis showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect, while X-ray analyses evidenced rise of bone density. Implantation of Ca/Co-Hap enables rapid new formation, thus becoming the material of choice for accelerated reduced bone density regeneration.

AB  - Systems for targeted delivery of drugs with high affinity for specific organs, tissues, and cells were introduced at the beginning of the twentieth century. Recently, this concept is attracting much attention. These systems have shown most effective in reducing adverse effects of drugs. In recent years, scientists have created a system of nanoparticles for delivery of vitamin D. Special attention is paid to the role of vitamin D in the division and differentiation of osteoblasts and thus, in its role in osteogenesis. In this regard it was interesting to observe the local effects of the active form of vitamin D3 in osteogenesis, as a part of the system for local delivery. These effects were observed on an experimental model. Three types of nanobiomaterial were implanted in the artificially made defects in rats’ mandibular bone. Six weeks after implantation of hydroxyapatite, hydroxyapatite-coated vitamin D3 and hydroxyapatite nanoparticles coated with vitamin D3 mixed with the polymer, the animals were sacrificed, and the samples of mandibular alveolar bone were taken. The samples were adequately prepared and subjected to histopathological analysis. The best results in regeneration of osteoporotic jaw bone were obtained in the experimental group of animals where the implant was hydroxyapatite-coated vitamin D3 mixed with the polymer. The results show that nanobiomaterials with local delivery of active form of vitamin D3, used in this study, may be materials of the future in the area of weakened jaw bone tissue regeneration.

AB  - This study examined the role of Ca / Co-HAp nanoparticles on time distance in regeneration of osteoporotic alveolar bone in rats by biochemical blood markers analysis (ALP, Ca, Mg, P) and through histochemical analysis. The research was carried out on female Westar rats, aged 6-8 weeks. The obtained results for the biochemical blood markers showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect as early as in the mineralized tissues. It can be concluded that Ca/Co-HAp stimulates the regeneration of osteoporotic alveolar bone in tested animals as confirmed by the increased levels of biochemical blood markers and through histochemical analysis. Therefore, it can be concluded that Ca/Co-HAp nanocomposite should be choise material in the osteoconstructive processes in the future.

AB  - Antimicrobial materials based on hydroxyapatite are potentially attractive in a wide variety of medical and stomatological applications. The objective of this paper is to examine antimicrobic activity of cobalt-substituted calcium hydroxyapatite nanopowders and biphased calciumphosphate/ poli-lactide-co-glicolide. The antimicrobial effects of these substances (powders) against two pathogen bacterial strains- Escherichia coli (ATCC 25922) and Staphylococcus aureus (ATCC 25923) were tested by disc diffusion method and quantitative antimicrobial test in liquid medium. It was noted that the inhibition zone of the bacterial cells S. aureus around the sample of the Ca/Co- HAp, was a lot bigger compared to the inhibition zone of bacterial cells E. coli around the sample of the mentioned biomaterial, which means that this material has bigger antimicrobic activity on S. aureus, in relation to E. coli. Quantitative antimicrobial test in liquid medium demonstrate that cobalt-substituted calcium hydroxyapatite samples show viable cells reduction of both tested microorganisms. It may be concluded that nanoparticles of cobalt-substituted calcium hydoxyapatite nano-powders has a satisfactory antimicrobic activity according to the tested bacteria strain.

AB  - One of the prerequisites for a good prosthetic rehabilitation is well preserved bone tissue. However, osteoporosis as a modern age disease often compromises a good dental treatment. Oral implantology offers a solution through the use of various biomaterials. Most researchers focus on the synthesis of the nanomaterials with magnetic and paramagnetic properties. The aim of this study is to examine the role of Ca / Co-HAp nanoparticles in the regeneration of the osteoporotic alveolar bone in experimental animals by analyzing the biochemical blood markers (ALP, Ca, Mg, and P) and through histochemical analysis. The research was carried out on female Westar rats, aged 6-8 weeks. The obtained results for the biochemical blood markers showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect as early as in the 6th week of the experiment as well as an increased alkaline phosphatase activity in the mineralized tissues. The implantation of the biomaterials facilitates osteogenesis, justifying their use in the accelerated regeneration treatment of the osteoporotic alveolar bone.

abstract = "One of the prerequisites for a good prosthetic rehabilitation is well preserved bone tissue. However, osteoporosis as a modern age disease often compromises a good dental treatment. Oral implantology offers a solution through the use of various biomaterials. Most researchers focus on the synthesis of the nanomaterials with magnetic and paramagnetic properties. The aim of this study is to examine the role of Ca / Co-HAp nanoparticles in the regeneration of the osteoporotic alveolar bone in experimental animals by analyzing the biochemical blood markers (ALP, Ca, Mg, and P) and through histochemical analysis. The research was carried out on female Westar rats, aged 6-8 weeks. The obtained results for the biochemical blood markers showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect as early as in the 6th week of the experiment as well as an increased alkaline phosphatase activity in the mineralized tissues. The implantation of the biomaterials facilitates osteogenesis, justifying their use in the accelerated regeneration treatment of the osteoporotic alveolar bone.",

AB  - In this article, synthesis and application of calcium phosphate/poly-DL- lactide-co-glycolide (CP/PLGA) composite biomaterial in particulate form, in which each CP granule/particle is coated with PLGA, are described. Two types of the particulate material having different particle sizes were synthesized: one with an average particle diameter between 150 and 250 μm (micron-sized particles, MPs) and the other with an average particle diameter smaller than 50 nm (nanoparticles, NPs). A comparative in vivo analysis was done by reconstructing defects in osteoporotic alveolar bones using both composites. The material, CP granules/particles covered with polymer, was characterized using X-ray structural analysis, scanning electron microscopy, and atomic force microscopy. Changes in reparatory functions of tissues affected by osteoporosis were examined in mice in vivo, using these two kinds of composite materials, with and without autologous plasma. Having defined the target segment, histomorphometric parameters - bone area fraction, area, and mean density - were determined. The best results in the regeneration and recuperation of alveolar bone damaged by osteoporosis were achieved with the implantation of a mixture of nanoparticulate CP/PLGA composite and autologous plasma. After the implantation of microparticulate CP/PLGA, in the form of granules, mixed with autologous plasma, into an artificial defect in alveolar bone, new bone formation was also observed, although its formation rate was slower. © 2010 Wiley Periodicals, Inc.

AB  - In this study nanocomposite was used for reconstruction of alveolar bone defect of mandible. Specifically, the main interest for the use of magnetic nanoparticles in biomedical applications is that an inhomogeneous external magnetic field exerts a force on them, and thus they can be manipulated or transported to a specific diseased tissue by a magnetic field gradient. In addition, magnetic particles are of interest because they do not retain any magnetism after removal of the magnetic field. Specifically, inorganic biodegradable nanoparticles (including ceramics, like hydroxyapatite) will be functionalized with bioactive compounds that bond to bone of low mass. Extremely good results in the recovery of alveolar bone osteoporosis were achieved already after 6th week of the application of magnetic nanoparticles. After bonding specifically to osteoporotic bone and not healthy bone, magnetic nanoparticle systems will deliver bioactive compounds to locally increase bone mass. Implantation of magnetic nanoparticles will create bone construction and enable quick formation of new bone and become the material of choice for accelerated bone regeneration.

AB  - Istraživanje je sprovedeno sa ciljem da se dokaže aktivnost alkalne fosfataze, lokalizovane u dekalcifikovanoj kosti, uzete sa leve strane mandibule pacova, u kojoj je arteficijalno napravljen defekt i implantiran nano kalcijum-kobalt hidroksiapatit. Studija je obuhvatila 48 životinja, starosti 6-8 nedelja. Posle šest i dvadeset četiri nedelje od implantacije biomaterijala, uzeti su uzorci za histohemijsku analizu i fiksirani u PLP fiksativu. Zatim su dekalcifikovani, ukalupljeni u parafinski blok i obrađivani za svetlosno-mikroskopska ispitivanja i histohemijsko dokazivanje aktivnosti alkalne fosfataze. Histološka analiza je pokazala visok stepen reparatornih sposobnosti ugrađenog biokompozita u koštani defekat. Histohemijskom analizom je dokazana aktivnost alkalne fosfataze u mineralizovanom tkivu.

AB  - Osteoporoza kao bolest savremene civilizacije postaje sve aktuelnija u stomatologiji. Jedan od trendova u nalaženju rešenja je u oralnoj implantologiji kroz primenu biomaterijala sa magnetnim česticama. U ovoj studiji ispitivan je uticaj nanočestica Ca/Co-HAp na regeneraciju osteoporotične alveolarne kosti eksperimentalnih životinja analizom biohemijskih markera krvi (Ca, Mg, P). Istraživanje je sprovedeno na 48 Wistar pacovima, starosti 6-8 nedelja kojima je implantiran biometrijal u osteoprotičnoj alveolarnoj kosti. Dobri rezultati regeneracije osteoporotičnog tkiva su postignuti 6 nedelja nakon implantacije nanočestica. Biohemijski parametri koštane sinteze su u statistički značajnom porastu. Implantacija biomaterijala omogućava brzo formiranje nove kosti što ih čini materijalima izbora za ubrzanu regeneraciju kosti.

AB  - Bone tissue reconstruction and reparation is a big challenge in medicine. Biocomposite materials based on hidroxyapatite are widely used in reparation of bone defects. Adult bone marrow-derived stem cells may be considered in two categories: hematopoietic stem cells (HSC) from the bone marrow and mesenchymal stem cells from the bone marrow stroma (BMSC). HSC and BMSC do not only coexist in one organ, but functionally cooperate. BMSC have a critical role in the formation of hematopoietic microenvironment (HME). The aim of this study was to investigate the interactions between bone marrow cells and biocomposites based on HAp/PLLA (hidroxyapatite/poly-L-lactide) after subcutaneous implantation in Balb/c mice. In that purpose, bone marrow cells of Balb/c mice were seeded in HAp/PLLA tubes (15 mm×1,5 mm). The HAp/PLLA tubes with BMC was subcutaneously implanted with a needle into the intrascapsular region of the mouse. Implants were extracted after 2, 6 and 12 weeks. In implants after 2 and 6 weeks we found angiogenesis, collagenogenesis and new bone. Ectopic hematopoiesis was seen in implants after 12 weeks from implantation. As a good scaffold in the role of supporting osteogenesis and hematopoiesis, biocomposites HAp/PLLA can be good bone substitute materials in the bone reparation process. These results showed that the HAp/PLLA scaffold owned biological properties comparable to natural bone.
AB  - Ispitivana je biofunkcionalnost biokompozita hidroksiapatit/poli-L-laktida (HAp/ /PLLA) kao matrice za osteogenezu i hematopoezu posle subkutane implantacije singenim miševima Balb/c. HAp/PLLA oblikovan je u cevčice (15 mm×1,5 mm) koje su napunjene ćelijama koštane srži miša, a zatim implantirane. Kontrole su bile prokuvane tibije i femuri napunjene na isti način. Implanti su ekstrahovani posle 2, 6 i 12 nedelja i analizirani patohistološki. Angiogeneza i osteogeneza se vide već posle dve nedelje na implantima, što je izraženije na kostima. Vrlo izražena hematopoeza u implantima oba tipa se vidi posle dvanaest nedelja. HAp/PLLA može poslužiti kao matrica za ektopičnu osteogenezu i hematopoezu.

AB  - The need for bone graft materials in osteoreparation is tremendous. Many researches have shown that calcium-phosphate bioceramics have good biocompatibility and osteoconductivity. We used nanocomposite biomaterial calcium phosphate coated with poly (dl-lactide-co-glycolide) or N-CP/DLPLG. The goal of this investigation was to examine weather N-CP/DLPLG has ability to sustain growth of bone marrow cells after subcutaneous implantation in Balb/c mice. For that purpose N-CP/DLPLG implants with and without bone marrow cells (control) were made. Implants were extracted after eight days and eight weeks. In implants loaded with bone marrow cells after eight days and eight weeks we observed fields rich in cells, angiogenesis and collagen genesis. These results showed that N-CP/DLPLG has property of tissue scaffold which sustain bone marrow cells growth and collagen production. This represents a good way for further examination of N-CP/DLPLG as potentional tissue scaffold in osteoreparation.

Implanti su napravljeni od nanomaterijala mešanjem sa razblaženom krvlju i kostnom srži, a ekstrahovani su nakon osam dana i osam nedelja. Implanti bez kostne srži su bili kontrola. Nakon osam dana se na periferiji implanta vide gusta polja sa ćelijama, a u materijalu se vidi lakunarna organizacija sa prisustvom ćelija. Uočljiva je vaskularizacija i registrovan je kolagen. Na kontrolnim implantima je celularnost vrlo slaba. Posle osam nedelja redukovana je površina nanomaterijala i celularnost implanata.

AB  - Siloxane polimers have been widely used in biomedicine and pharmacy due to their biocompatibility. Hydroxyapatite (HAp) is a natural constituent of bones, and therefore widely used in maxillofacial and orthopedic surgery. HAp itself is amorphous and without elasticity, so its characteristics can be improved when combined with organic polymers. We evaluated the interaction of cells and composites made of polydimethylsiloxane (PDMS) and HAp by scanning electron microscopy (SEM) 10 days after their intraperitoneal implantation into Balb/c mice. Two composites which were different in the quantity of HAp were analyzed. Both of them showed high adhesive characteristics for different cell types. The erythrocytes in cell clusters could be seen on the surface of the composite with higher quantity of HAp.
AB  - Siloksanski polimeri široko se koriste u biomedicini i farmaciji. Hidroksiapatit (HAp) je prirodni konstituent kosti i zato je u širokoj upotrebi u maksilofacijalnoj i ortopedskoj hirurgiji. Sam HAp je amorfan i nema elastičnosti, pa se njegove osobine poboljšavaju kombinovanjem sa polimerima. Ispitivali smo interakciju ćelija i kompozita izrađenog od polidimetilsiloksana (PDMS) i HAp-a skening elektronskom mikroskopijom (SEM) 10 dana posle intraperitonealne implantacije ovih materijala u Balb/c miševe. Analizirana su dva kompozita koji se razlikuju u količini HAp-a. Oba se karakterišu visokom adhezivnošću za različite tipove ćelija. Eritrociti se vide u ćelijskim klasterima na površini kompozita sa većom količinom HAp-a.

AB  - Experimental and clinical studies have shown that HAp granules and powder may be successfully applied in reconstruction of bone defects. Composite HAp/PLLA biomaterial embodies good characteristics of each of these biomaterials. Differences in porosity, microstructure, compressive consistency as well as in bioresorbility of HAp/PLLA are achieved by using PLLA of different mole masses. The aim of this study was to analyse light microscopy of the tissue reaction and changes of implants made of combination of syngeneic bone fragments and HAp/PLLA biocomposites with PLLA molecule mass of 430 000 (HP2) and 50 000 (HP3). The mice were divided into 4 experimental studies, 8 animals each. Two groups underwent implantation one group of mice was falsely operated, and one group was used for general control. Pathohistological analysis of implants was performed three months after subcutaneous implantation. Phagocytosis of HAp/ PLLA and bone was noticed on both implants’ preparations. Blood vessels were also noticed confirming the process of angiogenesis around the implants’ particles. The holes around particles of biocomposites and bone fragments represented the signs of their resorption. The presence of multinuclear cells between implants’ particles was observed and it was more prominent in HP3 implants. Osteogenesis is more intense in HP3 and ectopic hematopoesis is present. Our results show that designed HAp/PLLA biocomposites have good biocompatibility, their combination with bone fragments of a vital bone enables osteogenesis and hematopoesis and these processes are more prominent if PLLA biocomposite with lower molecular mass was used.

Analysis of in vivo substitution of bone tissue by HAp/PLLA composite biomaterial with PLLA of different molecular weights using FTIR spectroscopy

AB  - Hydroxyapatite/poly-L-lactide (HAp/PLLA) composite biomaterial with PLLA of 50,000 and 430,000 g/mole molecular weight was studied in vivo. The biocomposite with PLLA of both molecular weights was implanted into mice and after 1 and 3 weeks removed from their organisms and analyzed by the FT-IR spectroscopy. After one week of testing in vivo, the implanted samples gave spectra in which absorption bands arising from new-formed functional groups of amine and peptide can be seen. Analysis of the spectra revealed faster formation of peptide compounds when the biocomposite with PLLA of lower molecular weight was used. After 3 weeks, the spectra of the biocomposite of both compositions were registered with pronounced absorption bands at about 3420 and 1650 cm-1 assigned to new-generated collagen, a component of the extracellular connective-tissue matrix. Also, in the case of the biocomposite sample with PLLA of lower molecular weight, disappearance of the previously present absorption band at about 1760 cm-1 originating from the C double bond O group of PLLA indicates complete bioresorption of the PLLA used. Analysis of the microstructures of the sample surfaces by scanning electron microscopy before and after implantation revealed bioresorption of the PLLA polymer phase in the system with PLLA of lower molecular weight and generation of collagen fibers at the sites of implanted bioresorptive PLLA. A mixture of autologous bone powder and HAp/PLLA biocomposite was also examined. After implantation, the same final products as in the case of HAp/PLLA composite biomaterial alone were found.
