Successful Preservation of Helichrysum plicatum L. Flowers Extract Using Novel Carriers by Spray Drying Method

AB  - Research hypothesis: Everlasting flowers (genus Helichrysum) represent a significant source of pharmacologically active secondary metabolites (flavonoids naringenin, kaempferol, apigenin) related to proven spasmolytic, antioxidant, and antimicrobial activity. A critical point in development of polyphenol rich extracts of H. plicatum is their limited stability, which can be solved using microencapsulation technique spray drying. Method : Helichrysum flower extract was spray dried using four different carriers, and their combinations. Besides the conventional ones (maltodextrin-MD, whey protein-WP, 20%, w/w), innovative carriers, cyclodextrins (beta-cyclodextrin-BCD, hydroxy-propyl-betacyclodextrin- HPCD, 15%, w/w) have been proposed to overcome the extract limitations. The liquid feed was spray dried in a Labtex ESDTi spray dryer, under following conditions: inlet 130  } 5°C and outlet 70  } 5°C temperatures, spraying air flow rate (75 m3/h), liquid feed (11 mL/min rate), atomization pressure (2 bar). The spray-dried Helichrysum extract (HE) was obtained and used for further analyses: total polyphenols and flavonoids contents, spectrophotometrically and individual components by HPLC method. The dried extracts were stored in brown glass tubes for 6 months under room temperature in order to determine changes in the content of total and individual compounds during period of real storage. Samples were placed at in stability chamber (Memmert, Schwabach, Germany), in the absence of light, during one month in order to determine the effect of storage during accelerated stability test. All experiments were executed in triplicates. One-way ANOVA was conducted to test the individual factors AND Duncan post hoc test for differences between the mean values detection (STATISTICA v.7.0.3, MS Office Excel v. 2010). Results : Obtained powders manifested high encapsulation efficiency (more than 80%), confirming spray drying as adequate microencapsulation technique. Spray dried HE, without a carrier addition, exhibited 97.32% and powders microencapsulated using carriers ranged from 80.07 (HE+HPCD+MD) to 96.45% (HE+HPCD) of EE%, suggesting HEs active compounds are successfully microencapsulated into examined carriers. Spray-dried HE exhibited 106.32 mg GAE/g of polyphenol content, and powders produced using different polymers ranged from 83.25 (HE+HPCD+WP) to 100.28 mg GAE/g (HE+HPCD). The highest total flavonoids content was achieved in spray-dried HE. Among powders obtained using carriers, total flavonoids ranged from 15.66 (HE+HPCD+WP) to 21.55 (HE+HPCD) mg catechin/g. HPCD complexes exhibited the highest polyphenols and flavonoids content. Results of HPLC method confirmed that mainly presented flavonoids in obtained powders were naringenin, kaempferol, quercitrin, isoquercitrin, apigenin, and apigenin and naringenin derivates. Kaempferol-3-O-glycoside was the most dominant compound presented in all tested samples. Analysis of total polyphenols, flavonoids and individual compounds were carried after one month of accelerated and 6 months of real storage conditions. After one-month, total polyphenols and flavonoids increased in all examined samples. During the accelerated storage, water migrate from CDs and other carrier complexes, resulting the higher concentration. After 6 months, total polyphenols and flavonoids increased compared to the contents before storage tests, while decreased compared to 30 days without statistical significance. Content of individual compounds did not decrease significantly after 6 months, indicating that process of spray drying was suitable for stabilization of HE active compounds and indicating good preservation using polymers. Discussion : Spray drying of Helichrysum flower extract to obtain powders with high retention of bioactive compounds was evaluated. All microencapsulated bioactive principles reached high content and preservation during storage, based on good preservation. This could be important for further use in pharmaceutical and food industry due to its confirmed health benefits.

AB  - Roots of the willow gentian (Gentiana asclepiadea L., Gentianaceae) are used in folk medicine to treat gastrointestinal diseases.Spray drying is the most commonly used technique for microencapsulation of plant extracts in the pharmaceutical, cosmetic and food industries. The choosing of suitable carriers is one of the critical points in the development of microencapsulated powder with the desired physicochemical properties.The extract of the willow gentian roots was spray-dried using different carriers (maltodextrin (MD) and whey protein (WP)at concentrations of 20, 40, and 60%; pectin (P), starch (S), and gelatin (G) at concentrations of 2.5, 5, and 7.5%).The aim of this study was to examine the ability of the obtained powdersto dissolveand absorb water.For this purpose, the values of water solubility (WSI) and water absorption index (WAI) were determined.WSI is a measure of the powder degradation and dissolving in water, while WAI represents an indicator of water absorption ability. It is desirable that instant tea powders show high WSI and low WAI values.WSI values ranged from 71.15% (7.5% P) to 91.94% (7.5% G) indicating good powder disintegration properties. Concentration-dependent decrease in WSI values was observed for WP, P and S, while the opposite trend was observed in the case of MD.The carrier-free powdershowed relatively low WAI value (13.32%). Powders with 20% MD and 5% S showed slightly lower WAI values (12.77 and 12.72%, respectively). On the other hand, powders with 5 and 7.5% P showed remarkably high WAI values of 91.59 and 91.49%, respectively. A concentration-dependent increase in WAI was observed in powders encapsulated withMD and WP.All encapsulated powders showed acceptable disintegration properties (high WSI), while in terms of water absorption (low WAI) powders with 20% MD and 5% S were the most suitable.

AB  - Willow gentian as a source of bitter compounds is traditionally used for digestive disorders. Gentiana root extract was spray dried using five different carriers (maltodextrin, whey protein, pectin, starch, gelatine) at various concentrations. Powders were characterized in terms of physical properties and encapsulation efficiency of bioactive compounds. The moisture content of all powders was between 1.78 and 3.46 %, and bulk density from 0.23 to 0.32 g/mL. Powders produced with maltodextrin and whey protein provided the highest yield (around 75 and 70 %, respectively) and the lowest hygroscopicity (6 and 7%, respectively). Gelatin and pectin provided powders with the highest encapsulation efficiency of total phenolic as well as the individual compounds. The stability of encapsulated bioactive compounds was studied after 6 months, and the most stable in all samples were gentiopicrin and sweroside with their content decreased by 10 % only. This study has shown that spray drying of gentian root extract produces powders with good physical properties and encapsulation of bioactives.

AB  - This paper deals with development of a personal robot functioning as a part of a smart home. The overall system is based on the cloud computing model to leverage processing, data sensing and communication power of the robot. At the semantic level, it provides capabilities for improved perceiving and expressing the emotions based on analysis of collected and exchanged history data on emotional states and actions.

AB  - The paper is addressed to prototyping of technology platform aimed to develop of ambient-aware human-centric indoor service robot with attributes of emotional intelligence to enhance interaction with social environment. The robot consists of a wheel-based mobile platform with spinal (segmented) torso, bi-manual manipulation system with multi-finger robot hands and robot head. Robot prototype was designed to see, hear, speak and use its multimodal interface for enhanced communication with humans. Robot is capable of demonstrating its affective and social behavior by using audio and video interface as well as body gestures. Robot is equipped with advanced perceptive system based on heterogeneous sensorial system, including laser range finder, ultrasonic distance sensors and proximity detectors, 3-axis inertial sensor (accelerometer and gyroscope), stereo vision system, 2 wide-range microphones, and 2 loudspeakers. The device is foreseen to operate autonomously but it may be also operated remotely from a host computer through wireless communication link as well as by use of a smart-phone based on advanced client-server architecture. Robot prototype has embedded attributes of artificial intelligence and utilizes advanced cognitive capabilities such as spatial reasoning, obstacle and collision avoidance, simultaneous localization and mapping, etc. Robot is designed in a manner to enable uploading of new or changing existing algorithms of emotional intelligence that should provide to robot human-like affective and social behavior. The key objective of the project presented in the paper regards to building advanced technology platform for research and development of personal robots aimed to use for different purpose, e.g. robot-entertainer, battler, robot for medical care, security robot, etc. In a word, the designed technology platform is expected to help in development human-centered service robots to be used at home, in the office, public institutions, etc.

AB  - This paper describes a prototype of an ambient-intelligent advanced service robot of anthropomorphic characteristics that is intended for operation in indoor environment as well as for safe interaction with people. The robot consists of a wheeled mobile platform with a spinal (segmented) torso, bi-manual manipulation system with hands, and a robot head with capabilities to see, hear and speak. It is equipped with a number of advanced sensors, including indoor laser range finder, several ultrasonic probes as distance sensors and obstacle detectors, 3-axis inertial sensors with gyroscope, stereo vision system, 2 wide-range microphones, and 2 speakerphones. Its operation is autonomous but it may be controlled from a host computer through a wireless link. The robot prototype is expected to express advanced cognitive capabilities including spatial understanding, autonomous motion, affective and social behavior. The development of the robot is a joint effort of four Serbian academic institutions and it is expected to have it fully operational in the second half of 2015.
