AB - In the field of oral implantology the loss of bone tissue prevents adequate patient care, and calls for the use of synthetic biomaterials with properties that resemble natural bone. Special attention is paid to the risk of infection after the implantation of these materials. Studies have suggested that some nanocontructs containing metal ions have antimicrobial properties. The aim of this study was to examine the antimicrobial and hemolytic activity of cobalt-substituted hydroxyapatite nanoparticles, compared to hydroxyapatite and hydroxyapatite/poly-lactide-co-glycolide. The antibacterial effects of these powders were tested against two pathogenic bacterial strains: Escherichia coi (ATCC 25922) and Staphylococcus aureus (ATCC 25923), using the disc diffusion method and the quantitative antimicrobial test in a liquid medium. The quantitative antimicrobial test showed that all of the tested biomaterials have some antibacterial properties. The effects of both tests were more prominent in case of S. aureus than in E coli. A higher percentage of cobalt in the crystal structure of cobalt-substituted hydroxyapatite nanoparticles led to an increased antimicrobial activity. All of the presented biomaterial samples were found to be non-hemolytic. Having in mind that the tested of cobalt-substituted hydroxyapatite (Ca/Co-HAp) material in given concentrations shows good hemocompatibility and antimicrobial effects, along with its previously studied biological properties, the conclusion can be reached that it is a potential candidate that could substitute calcium hydroxyapatite as the material of choice for use in bone tissue engineering and clinical practices in orthopedic, oral and maxillofacial surgery.

AB - Composite biomaterials comprising nanostructured hydroxyapatite (HAp) have an enormous potential for natural bone tissue reparation, filling and augmentation. Chitosan (Ch) as a naturally derived polymer has many physicochemical and biological properties that make it an attractive material for use in bone tissue engineering. On the other hand, poly-D,L-lactide-co-glycolide (PLGA) is a synthetic polymer with a long history of use in sustained drug delivery and tissue engineering. However, while chitosan can disrupt the cell membrane integrity and may induce blood thrombosis, PLGA releases acidic byproducts that may cause tissue inflammation and interfere with the healing process. One of the strategies to improve the biocompatibility of Ch and PLGA is to combine them with compounds that exhibit complementary properties. In this study we present the synthesis and characterization, as well as in vitro and in vivo analyses of a nanoparticulate form of HAp coated with two different polymeric systems: (a) Ch and (b) a Ch-PLGA polymer blend. Solvent/non-solvent precipitation and freeze-drying were used for synthesis and processing, respectively, whereas thermogravimetry coupled with mass spectrometry was used for phase identification purposes in the coating process. HAp/Ch composite particles exhibited the highest antimicrobial activity against all four microbial strains tested in this work, but after the reconstruction of the bone defect they also caused inflammatory reactions in the newly formed tissue where the defect had lain. Coating HAp with a polymeric blend composed of Ch and PLGA led to a decrease in the reactivity and antimicrobial activity of the composite particles, but also to an increase in the quality of the newly formed bone tissue in the reconstructed defect area.

AB - Composite biomaterials based on nano-hydroxyapatite have an enormous potential for natural bone tissue reparation, filling and augmentation. Multifunctional nanoparticulate systems based on HAp coated with biocompatible and bioresorbable polymers make a separate group of filler systems in bone tissue engineering [1,2]. Chitosan has many physicochemical (reactive OH and NH2 groups) and biological (biocompatible, biodegradable) properties that make it an attractive material for use in bone tissue engineering. However, chitosan may induce thrombosis and it is therefore unsuitable as blood – contacting biomaterial. One of the strategies to improve the biocompatibility of chitosan is combination of this biopolymer with compounds that exhibit complementary properties.
In our studies, we present the synthesis, characterization, in vitro and in vivo research of a particulate form of nano HAp-coated polymer systems. We synthesized nanoparticulate HAp coated with chitosan (Ch) and a chitosan-poly-D,L-lactide-co-glycolide (Ch-PLGA) polymer blend obtained via the solvent/non-solvent method and freeze-drying processing. We also examined the possibility of using Thermo-Gravimetric Analysis/Differential-Thermal Analysis (DTA/TGA) coupled on-line with mass spectrometry (MS) as a finger print for identification purposes in coating processes. The quantitative antimicrobial test has shown that HAp/Ch-PLGA have some antibacterial properties (MIC (mg/mL): Pseudomonas aeruginosa – 6.40, Staphylococcus aureus – 6.40, Staphylococcus epidermidis – 3.20). MTT assay was used to test cytotoxicity and cell viability. By using HAp/Ch-PLGA in the form of a filler a high level of reparatory ability, with the presence Haversian canals and cement lines in reconstructed of bone defect, was achieved in vivo.

AB - Hydroxyapatite (HAp) is an extensively studied material with known biocompatible and osteoconductive properties in bone tissue reconstruction. The improvement of the osteogenetic potential of HAp has been tested through modification of its structure, by replacing Ca2+ ions with Co2+ ions. In our study, we comparatively analyze the osteogenetic potential of the synthesized HAp and Co2+-substituted HAp (HAp/Co) designed on the nano-scale with the aim of specifically stimulating osteogenesis in vivo. We present a quantitative study of the microscopic organization and structure of the newly formed tissue in a bone defect after 12 weeks and 24 weeks. A quantitative analysis of the calcium, magnesium and phosphorus content in the defect and its close environment was used to determine the deposition of minerals after bone reconstruction. The defect reconstructed with HAp/Co nanoparticles (Co2+ content 12 wt%) was filled with a new tissue matrix composed of dense collagen fibers containing centers of mineralization after 24 weeks. The mineral deposition rate was also higher when the defect was reconstructed with HAp/Co than when it was filled with pure HAp. A histological analysis confirmed that the alveolar bone, in which osteoporosis-induced defects were repaired using HAp/Co nanoparticles, was recuperated. © 2015 Jilin University.

abstract = "Hydroxyapatite (HAp) is an extensively studied material with known biocompatible and osteoconductive properties in bone tissue reconstruction. AB - The influence of glucocorticoids on the composition and mineral/organic content of the mandible in tested animals after recovery and healing phase was investigated in this work. The results of FTIR analysis demonstrated that bone tissue composition was changed after glucocorticoid treatment. The increase of calcium, magnesium, phosphorus content and mineral part of bones was statistically significant in recovery phase and in treatment phase that included calcitonin and thymus extract. Some changes also happened in the organic part of the matrix, as indicated by intensity changes for already present IR bands and the appearance of new IR bands in the region 3500–1300 cm−1.

AB - Hydroxyapatite (HAp) nanoparticles coated with bioresorbable polymers have been successfully used as filler, carriers of antibiotics, vitamins and stem cells in bone tissue engineering. Interactions in the microenvironment of nanoparticulate hydroxyapatites and tissues are of a particular interest and present a challenge in regenerative medicine. The phenomena that influence the success of targeting and controlled operation or therapeutic nanosystems are numerous and complex: size, shape, surface charge, functional groups of nanoparticles, electrical double-layer formation, zeta potential, partial molar free energy, sorption molar free energy, isoelectric point of solid–liquid interface; receptor–ligand binding interactions of nano-bio interface etc. In our studies we present the synthesis, characterization, in vitro and in vivo research of nanoparticulate form of HAp-coated polymers systems. Synthesized nanoparticulate HAp coated with different types of bioresorbable polymers: poly(D,L-lactide-co-glycolide), chitozan and other similar by the solvent/non-solvent method. The physical and chemical analyses have confirmed that HAp particles are coated with bioresorbable polymers. Results of spectroscopic analysis suggests formation of hydrogen bonds between the surface groups of HAp and =O and –H groups in the polymer chain from bioresorbable polymers. By adding nanoparticulate HAp in the polymer matrix was achieved the changes in the partial molar free energy and sorption molar free energy of the system. In vitro results showed satisfactory biocompatibility of nanoparticulate HAp-coated polymers systems. The use of these systems in the form of filler was achieved a high quality osteogenesis in reconstruction of bone tissue, in vivo. The biodistribution of 125I-labeled HAp particles after intravenous injection in rats showed the potential use of these materials as a organ-targeting system: uncoated-HAp mostly targeted the liver, coated-HAp spleen and lungs.

AB - Recent studies have shown that the behavior of nanoparticles in in vivo conditions is not solely dependant on their physical properties, such are size and shape. The surface of nanoparticles in biological fluids interacts with biomolecules such are proteins, adsorbs these molecules, and leads to formation of nanoparticle-biomoelcular complex known as “protein corona”. This protein corona changes the properties of nanoparticles and their behavior in vivo. In this work three types of nanomaterial based on Hap and polymers were incubated in rats plasma, and the identification and quantification of proteins in protein corona that had formed around these nanoparticles, was done by means of electrophoresis and mass spectrophotometery.

AB - The loss of teeth and damage to the jaw bone occur under different circumstances. The deficit may be the result of wearing inadequate dentures, residual ridge resorption, osteoporosis, periodontal disease, trauma, tumors, radiation, etc. The deficit of bone tissue can be a substantial problem because it prevents adequate prosthetic patient care and restoration of disturbed functions of stomatognathic system. With such patients, there is often a need for preprosthetic surgical procedure through the application of various techniques and materials in order to resolve aftermentioned bone shortcomings. The most appropriate biological materials for the reconstruction are autogenous bone graft and artificial bone. With the development of technology and specific
biomaterials it is tried to avoid situations that require autogenous bone grafting, since it involvs long surgical procedure with the possibility of postoperative complications. The application of synthetic biomaterials, whose properties are similar to natural hydroxyapatite, promotes the biological potential for bone tissue repair. Synthetic biomaterials have great biocompatibility and sterility and do not act antigenicaly. The disadvantages of this material are overcome by the addition of organic polymers. Thanks to the stability and flexibility of hydroxyapatite structure, a large number of ionic changes are possible, both cationic and anionic, which improve the characteristics of synthetic hydroxyapatite. Biomaterials that include different drugs, such as antibiotics, vitamins and other preparations, could be used for the treatment of complications after surgical procedures and infection of bone tissue. Our experimental studies indicate that biomaterials based on hydroxyapatite are valuable materials that can be used for preprosthetic surgical preparation.

AB - Indications exist that paramagnetic calcium phosphates may be able to promote regeneration of bone faster than their regular, diamagnetic counterparts. In this study, analyzed was the influence of paramagnetic cobalt-substituted hydroxyapatite nanoparticles on osteoporotic alveolar bone regeneration in rats. Simultaneously, biocompatibility of the material was tested in vitro, on osteoblastic MC3T3-E1 and epithelial Caco-2 cells in culture. The material was shown to be biocompatible and nontoxic when added to epithelial monolayers in vitro, while it caused a substantial decrease in the cell viability as well as deformation of the cytoskeleton and cell morphology when incubated with the osteoblastic cells. In the course of 6 months after the implantation of the material containing different amounts of cobalt, ranging from 5 to 12 wt%, in the osteoporotic alveolar bone of the lower jaw, the following parameters were investigated: histopathological parameters, alkaline phosphatase and alveolar bone density. The best result in terms of osteoporotic bone tissue regeneration was observed for hydroxyapatite nanoparticles with the largest content of cobalt ions. The histological analysis showed a high level of reparatory ability of the nanoparticulate material implanted in the bone defect, paralleled by a corresponding increase in the alveolar bone density. The combined effect of growth factors from autologous plasma admixed to cobalt-substituted hydroxyapatite was furthermore shown to have a crucial effect on the augmented osteoporotic bone regeneration upon the implantation of the biomaterial investigated in this study.

AB - Cholecalciferol, vitamin D3, plays an important role in bone metabolism by regulating extracellular levels of calcium. Presented here is a study on the effects of the local delivery of cholecalciferol (D3) using nanoparticulate carriers composed of hydroxyapatite (HAp) and poly(d,l-lactide-co-glycolide) (PLGA). Multifunctional nanoparticulate HAp-based powders were prepared for the purpose of: (a) either fast or sustained, local delivery of cholecalciferol, and (b) the secondary, osteoconductive and defect-filling effect of the carrier itself. Two types of HAp-based powders with particles of narrowly dispersed sizes in the nano range were prepared and tested in this study: HAp nanoparticles as direct cholecalciferol delivery agents and HAp nanoparticles coated with cholecalciferol-loaded poly(d,l)-lactide-co-glycolide (HAp/D3/PLGA).

Satisfying biocompatibility of particulate systems, when incubated in contact with MC3T3-E1 osteoblastic cells in vitro, was observed for HAp/D3/PLGA and pure HAp. In contrast, an extensively fast release of cholecalciferol from the system comprising HAp nanoparticles coated with cholecalciferol (HAp/D3) triggered necrosis of the osteoblastic cells in vitro. Artificial defects induced in the osteoporotic bone of the rat mandible were successfully reconstructed following implantation of cholecalciferol-coated HAp nanoparticles as well as those comprising HAp nanoparticles coated with cholecalciferol-loaded PLGA (HAp/D3/PLGA). The greatest levels of enhanced angiogenesis, vascularization, osteogenesis and bone structure differentiation were achieved upon the implementation of HAp/D3/PLGA systems.

AB - The lack of bone in the jaw tegmenta inflicts major problem and leads to an inability to adequately treat stomatoprosthetic patients. If the bone tissue damage is minor, the balanced activities of osteoblasts and bone osteoclasts can repair it independently. If the damage is bigger it is necessary to support the biological potential to repair the bone, and for that reason nanoparticle
biomaterial Ca / Co-HAp was used in this study. The research was done on rats with uniform anatomical and physiological characteristics. Assessment of repair and consolidation of the jaw bone tegmenta was performed by istomorphometric and SEM analysis. The best results were obtained in the experimental group of animals where the Ca / Co-Hap was mixed with autologous
plasma. Following the implementation of the above mentioned nanocomposites, a significant formation of new bone was evident on the SEM analysis, as well as the rising of histomorphometric parameters of bone formation, which indicates that the Ca / Co-HAp nanocomposite is the material of choice for the rapid regeneration and repair of bone jaw tegmenta.

AB - We have examined the effect of extracts of the calcium phosphate/poly(DL-lactide-co-glycolide) (CP/PLGA) and cobalt-exchanged hydroxyapatite (CoHAp), on the viability of Saos2 osteoblastlike cells. Extracts were prepared by incubation for 3 days at 37°C in the cultivation medium. Conductivity and pH value of extracts were measured before viability assay. Cell viability was estimated by MTT test performed after 24 h incubation of cells with various concentrations of extracts. Extract of CP/PLGA acted more cytotoxic on osteoblasts than the extract of CoHAp. This difference in the effect of examined extracts can be explained by their different pH value and conductivity.

AB - Specific group of controlled drug delivery systems in bone tissue engineering are multifunctional nanoparticle systems (MNPs) based on hydroxyapatite coated with drug loadedbioresorbable polymer. This study illustrates the possibility for controlled synthesis of multifunctional nanoparticulate forms based on hydroxyapatite as a system for local delivery of vitamin D3 and secondary delivery of defect filler hydroxyapatite. The results were two type of nanoparticle powder of controlled shapes, sizes and properties: hydroxyapatite nano particles as vitamin D3 carriers (HAp/D3) and vitamin D3-loaded hydroxyapatite coated with poly-D,Llactide-co-glycolide (HAp/D3/PLGA) with particle sizes of d50=68 nm and d50=71 nm, respectively. Simultaneously, biocompatibility of the materials was tested in vitro, on osteoblastic MC3T3-E1 and epithelial Caco-2 cells in culture. Biocompatibility tests carried out on cell cultures have shown intact monolayers of epithelial cells in contact with both materials and no negative effects on the cell viability. The aim of this work was application in living/human systems, and it has been tested in vivo; artificially made bone defects of a mandible damaged by osteoporosis have been reconstructed with both types of materials. The best results were achieved 24 weeks after implantation of vitamin D3-loaded hydroxyapatite coated with poly-D,L-lactide-co-glycolide. Accelerated angiogenesis, vascularization, osteogenesis and bone structure differentiation has been achieved in the presence of specific islet-like forms of ossification centers.

AB - Bone loss during the systemic osteoporosis has an important role in dentistry and medicine. The aim of the study was the application of bioactive micro and nanocomposite materials, alone, and in combination with autologous plasma in osteoporotic jaw bones of rats with artificially induced osteoporosis. The effect of these composites was measured by histomorphometric and atomic absorption spectrophotometric analysis. According to the best obtained results in regeneration and recovery of the resorbed alveolar bone, it can be concluded that nanocomposite combined with autologous plasma may be the material of choice to replace the osteoporotic damaged jaw bone.

AB - In recent years, calcium hydroxyapatite (HAp) and its substituents are increasingly used in dentistry and medicine. The influence of nanoparticles Ca / Co-HAp in strengthening weak osteoporotic bone jaw tegmenta was tested in an experimental model. The study was conducted on Wistar soy rats, aged 6-8 weeks. The biomaterial was implanted in the osteoporoticly weakened mandible of these animals. The best results in the strengthening of the lower jaw bone tegmenta were achieved 24 weeks after implantation of hydroxyapatite nanoparticles in which the calcium ion was substituted with 12% of cobalt ions. Histochemical parameters of bone synthesis were in a statistically significant increase. SEM analysis showed a high degree of osteogenetic ability of nano-particulate material implanted in the bone defect.

AB - The aim of this study was to compare the effects of two types of nanomaterials, cobalt-exchanged hydroxyapatite (CoHAp) and calcium phosphate/poly-(DL-lactide-co-glycolide) (CP/PLGA), on the viability of Saos-2 osteoblast-like cells, using MTT test. We have examined the suspension of this materials in the following concentrations: 1.6, 8, 40, 200, 1000 and 2500 μg/ml, as well as extracts in concentration range from 2,5% to 100%. Both materials showed cytotoxic effect at higher concentrations of suspension and extract, respectively, but they were not cytotoxic at lower concentrations. CP/PLGA acted stronger cytotoxic compared to CoHAp, regardless of whether it is examined suspension or extract. CoHAp in small concentrations of suspension and extract acted slightly stimulatory on cells. This suggests that CoHAp may have advantage for use in the in vivo systems.

AB - Huge range of tested biomaterials in recent decades has emerged as an ideal scaffold for cell growth, but few have demonstrated clinical efficacy. Among them, synthetic hydroxyapatite (HAp, Ca10(PO4)6(OH)2) is the most promising because of its biocompatibility, bioactivity, and osteoconductivity. Biocompatibility represents the primary concern for any material to be used as a substitute for natural tissue. Hydroxyapatite particles interact with numerous cellular systems in vivo, and some of these interactions may lead to cell damage and to stimulate platelet activation, coagulation and thrombus formation. The aim of this work was to examine the hemocompatibility of nanocalcium hydroxyapatite substituted with 5% and 12% cobalt (Ca /CoHAp) and hydroxyapatite/polylactidcoglicolid (HAp / PLGA) in relation to pure HAp by testing their hemolytic activities. The results show the discrepancy in hemolytic activity of implanted matherials. The degree of crystallinity of samples had a more dominant influence on hemolysis than the percentage of substituted cobalt. Hemolysis ratios of the nanocalcium hydroxyapatite substituted with cobalt samples were below 3%, indicating good blood compatibility and that they are promising for medical application.

abstract = "Huge range of tested biomaterials in recent decades has emerged as an ideal scaffold for cell growth, but few have demonstrated clinical efficacy. Hemolysis ratios of the nanocalcium hydroxyapatite substituted with cobalt samples were below 3%, indicating good blood compatibility and that they are promising for medical application.",

AB - One of the trends in the treatment of reduced bone density is in oral implantology through the use of new biomaterials with paramagnetic properties. In this in vivo study the effect of Ca /Co-HAp nanoparticles (calcium/cobalt-hydroxiapatite) in the regeneration of the reduced alveolar bone density in experimental animals was examined by analyzing both the biochemical and histochemical markers and through analysis of alveolar bone density. The research was carried out on female Westar rats, aged 6-8 weeks, whom have been implanted nanobiomaterial in reduced alveolar bone density. Good results in regeneration of reduced bone density were achieved in six weeks after implantation of the nanoparticles with paramagnetic properties. Biochemical markers of osteogenesis showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect, while X-ray analyses evidenced rise of bone density. Implantation of Ca/Co-Hap enables rapid new formation, thus becoming the material of choice for accelerated reduced bone density regeneration.

AB - Systems for targeted delivery of drugs with high affinity for specific organs, tissues, and cells were introduced at the beginning of the twentieth century. Recently, this concept is attracting much attention. These systems have shown most effective in reducing adverse effects of drugs. In recent years, scientists have created a system of nanoparticles for delivery of vitamin D. Special attention is paid to the role of vitamin D in the division and differentiation of osteoblasts and thus, in its role in osteogenesis. In this regard it was interesting to observe the local effects of the active form of vitamin D3 in osteogenesis, as a part of the system for local delivery. These effects were observed on an experimental model. Three types of nanobiomaterial were implanted in the artificially made defects in rats’ mandibular bone. Six weeks after implantation of hydroxyapatite, hydroxyapatite-coated vitamin D3 and hydroxyapatite nanoparticles coated with vitamin D3 mixed with the polymer, the animals were sacrificed, and the samples of mandibular alveolar bone were taken. The samples were adequately prepared and subjected to histopathological analysis. The best results in regeneration of osteoporotic jaw bone were obtained in the experimental group of animals where the implant was hydroxyapatite-coated vitamin D3 mixed with the polymer. The results show that nanobiomaterials with local delivery of active form of vitamin D3, used in this study, may be materials of the future in the area of weakened jaw bone tissue regeneration.

AB - Multifunctional drug delivery system based on hydroxyapatite may be a research challenge in the treatment and reconstruction of bone tissue. Vitamin D3 has a positive effect on osteogenesis as it increases osteoblast differentiation and mineralization of bone tissue. The purpose of the study presented in this paper has been to examine the possibility of the synthesis of a new multifunctional nanoparticulate system for local delivery of vitamin D3 suitable for applications in bone engineering. The synthesis, characterization and application of three nano particle systems was shown: hydroxyapatite, hydroxyapatite/vitamin D3 and hydroxyapatite/PLGA/vitamin D3. The obtained vitamin D3-loaded nanoparticles based on hydroxyapatite. Characterization was performed with wide-angle X-ray structural analysis (XRD), infrared spectroscopy (FT-IR), field-emission scanning electron microscopy (FESEM), zeta potential (ZP) and particle sized distribution (PSD) methods. Basic in vitro studies were performed implantation of materials in rats. XRD and FT-IR analyses confirmed that the vitamin D3 is loaded hydroxyapatite and PLGA. Different values of zeta potential may refer to different phenomenological processes during in vivo studies.

AB - This study examined the role of Ca / Co-HAp nanoparticles on time distance in regeneration of osteoporotic alveolar bone in rats by biochemical blood markers analysis (ALP, Ca, Mg, P) and through histochemical analysis. The research was carried out on female Westar rats, aged 6-8 weeks. The obtained results for the biochemical blood markers showed statistically significant rise. Histological analysis revealed high level reparatory skills of the biocomposite implanted in the bone defect as early as in the mineralized tissues. It can be concluded that Ca/Co-HAp stimulates the regeneration of osteoporotic alveolar bone in tested animals as confirmed by the increased levels of biochemical blood markers and through histochemical analysis. Therefore, it can be concluded that Ca/Co-HAp nanocomposite should be choise material in the osteoconstructive processes in the future.
